“Remarkably, individuals showed an average decrease in biological aging of 8 years”
This week in the journal Aging-US, we get a look at the first data of Rejuvant, an alpha ketoglutarate (AKG) based formulation, in human patients.
It was previously shown in a study that AKG supplementation increases the lifespan of the worms C.elegans and I reported on the study that came out last year showing that the calcium-salt of AKG extended both the lifespan and healthspan of mice.
In this publication they show 42 participants take Rejuvant and used DNA methylation (epigenetic age) to assess their "biological age". The data shows it conferred an average 8 year reduction in biological aging, after an average of 7 months of use. In this video we will break down this paper completely, address exactly what the data shows, what it doesn’t show and what I think the take homes are? We'll also discuss the limitations such as lack of clinical markers and no placebo group!!

I probably should've mentioned that a larger clinical trial is being conducted in Singapore using AKG and looking at more measurements (Prof Brian Kennedy) so hopefully we will get more data soon!

Interesting to note that the lines on the delta graph basically have two different slopes. One group saw moderate improvement the other saw much more pronounced improvement. Would be very interesting to see if a relevant variable could be identified to distinguish them.

Thanks for this video. My friend tried it but said it had 0 effect on his running speed or ability (as a functional indicator). Like myself and a lot of females, bodybuilding benefit isn't really of interest to me. I liked the fact you touched on methylation biomarkers as not necessarily indicators of true clinical functionality.

Also there is no control group to compare the measurements against, there should be a group where they measure before and after 7 months to see what the changes are in DNA methylation. There also needs to be peer reviewed, 42 is also not a big number although statistically it's significant enough to predict normally distributed behaviors to 95% confidence levels, but this is assuming aging improvements are normally distributed through AKG supplements. This also needs to be replicated multiple times as well, double blind test...this isn't rigorous enough for scientific publication.

There was another study recently that came to light that suggested that an 8 week healthy diet reduced epigenetic age by 1.7 years or so. Push the timeline forward that suggests that in 1 years of a healthy diet / lifestyle / supplement tweaks you become 10 years younger and in 4 years I would be a baby again. To be frank these claims a bordering on ridiculous and far too good to be true. They are crunching the data in a sensationalist way I don't buy it for one second. We can't move forward if we clutch hold of nonsense. I'm deeply suspicious of who funded the study and why it was conducted. I am happy to be wrong about this but I doubt anyone is gonna make my day anytime soon.

And it doesn't have to be some chemical concoction, you can take it right away too and have impressive results on skin, digestive health and increased cell apoptosis and anti inflammatory. With all the shortcomings considered (not placebo controled and non assement of some functional data being more serious, the others - I don't see them as important), I think this is an important study as it points to something else than NAD+ boosters. Thank you for this post.

The arginine salt of AKG is sold quite cheaply as a bodybuilding supplement. Can anyone think of a reason why this wouldn't be just as effective as the calcium salt?

Chasing a drug to make you younger as you decline right into your grave is not a good anti-aging strategy at the moment. Want to stave off old age? Exercise HARD, and eat clean.

Well done. I appreciate your approach of presenting the paper as objectively as possible, then providing logical critique. Your points were spot on, specifically the lack of control and the choice of methylation test. I am largely unfamiliar with attempts to connect methylation with aging, as I have focused most of my attention on methylation, environment, and inherited traits. Is there much data linking methylation and aging? Is it showing cause or coincidence?

ONE CONCERN: Is calcification of the arterial walls a possible danger with the Ca-AKG (Calcium Alpha-Ketoglutarate) supplements because there is a lot of caution with this possibly being the case with other calcium supplements?

Thanks Eleanor. Yes the placebo effect, which can be powerful, needs to be accounted for. Would like to see how this stacks up against the diet/lifestyle study done by Kara Fitzgerald., which seem to show age reversal over a much short intervention.

Must be difficult getting funding without some sort of commercial finance.
Interesting that potentially long term use, the body may get use to its affect.

Does simply changing the DNA methylation pattern truly reduce aging? The pattern is correlated with age, but the AKG intervention may itself change the correlation. This study is not at all convincing.

Very excited that this study was finally done on humans, very slow progress in this direction taking into account that similar study on nematode worms was done almost 10 years ago. I am wondering if longevity effects are related to the ability of α-Ketoglutaric acid to bind ammonia therefore helping body to detoxify metabolic byproducts.

I was listening to Peter Attia's podcast recently and was surprised to here him say where are no good biomarkers for aging. I know that dna methylation clocks aren't perfect, but I was under the impression that they were pretty good?! Also do you take a AKG supplement?

Been taking AAKG for mitochondrial benefits for almost two years. Probably too young to have noticable de-ageing effects but can confirm I feel and train orders of magnitude better in most all capacities

Is it not possible that AKG directly influences methylation? Were possible mechanisms for this proposed in the paper? After all, it seems much more plausible that a medication would directly affect the methylation sites that they happened to measure rather than literally reverse aging, which would mean having targeted and positive effects on a huge number of pathways, many more than would be required to just affect DNA methylation. Doesn't Occam's razor favor the latter hypothesis until proven otherwise?

Any word on how long it will take Brian Kennedy to confirm this? 8 years seems highly significant and one would think there would be an army of researchers attempting to prove or disprove, and also raising the Q why limit the research to the expensive calcium version?

What’s with the vitamin A and D? I thought men benefit from equal if not greater vitamin D levels and women don’t have vastly different vitamin A requirements from men.
Couldn’t the inclusion of either also compromise the findings since they could just be treating deficiencies or whatever. Hope I’m missing something and/or this isn’t the only formulation that’s been used in high-profile Ca-AKG research (though since Rejuvant keeps funding it…)

A healthy diet cutting out heavy drinking, overly rich foods and drugs in general will 'reduce' your age from a health standpoint too.
I highly doubt there is a wonder pill to replace a healthy lifestyle, just like the 'too good to be true' weight loss programs also require diet changes.
Taking supplements and changing to a healthy diet has always improved people's health.
Let's get to that stuff they used to make an old rat look young.

I'm considering starting to take MNM, TMG, and. AKG. Is there any worries that any of these will have an adverse effect on me. I had a bi-cuspid aortic valve replaced and I now take Warfarin. I can't find any information, and would like to make an informed decision before trying these.

I thought it was normal practice for pharma companies to fund studies for their products. It makes sense that they want the scientiric proof of claims as a selling point. If the results aren't the ones they want though, must they still publish and submit to peer review?

Okay, so this is odd. I read this comment about rejuvant from over a year ago on the age reversal network
"The speaker presented a talk indicating that users got substantial drop in DNA methylation age results - I seem to recall an average of 8 years after 6 months usage"
Either Rejuvant has been sitting on this data at least since 5/1/2020 or they just happened to repeat previous study results almost exactly. Or maybe it's totally normal to first publish results more than one year after a study has been completed?

Interesting, but all biomarkers of aging tests are not well tested, or compared or qualified - so this is not a ton of value. Did they use a bunch of different biomarker tests and just show the one that showed the best result?

I’ve been taking AAKG for about 10 months. I’m 65. Male. I’ve found I put on more muscle and less fat but the only side effect perhaps is my feet. I have pain on the outside of my right foot which I suspect is calcium deposits though I have not been to a doctor. I suspect the AAKG causes an accumulation of calcium in some way.

The only use of this study is to motivate other researches to do a proper placebo controlled study. As it stands now it's pure marketing so they can slap "according to research..." on their website and products.
