Thanks Dr. Sinclair .. a fan ... will be watching the outcome of your research ... since I am specializing in mitochondrial dysfunction pertaining to aging from a dietary approach, since as you know we have longevity mechanisms that repair all damage ... we just have to get out of there way and support our own metabolic processes as these bodily mechanisms are far more powerful than any supplement you can take ...,
70 Going On 100 … the Centenarian Diet … maybe 70 Going On 128 … the Hayflick Limit, or if a fan of Ray Kurzweil … then this is all a Moot Point.

Dr. Sinclair looks younger now (2022) than he did in 2014. That means his approach worked, i.e. diet, intermittent fasting, supplements, etc.

Wouldn't it be possible to look at the mitochondrial bacteria types which are living outside of human bodies, where they are less protected, to find some gens which could be used to make our Mitochondria more resilient to damage and mutations?

What determines the actual number of mitochondria per cell? Is it tissue dependent or based dynamically on energy needs?
Bruce Hay on the HumanOS podcast, mentions with respect to mitochondrial genomes, you have hundreds to thousands of them:
"In every cell we have hundreds to thousands of these organelles floating in the cytoplasm. They’re always being generated, and they have their own genomes. It’s not just one genome for all mitochondria, but thousands."
Does he mean that each mitochondria only has a portion of the mtDna? Isn't the mtdna so small (37 genes) that it barely codes for proteins anyways (13)?
He goes on to say, that the "nuclear DNA is well positioned for repair, because we have two copies of all of our chromosomes. What that means is, that if the copy you inherited from your father is damaged in some way, the copy you have from your mother can actually act as a template for repair. In that way, through homology based repair, you can oftentimes reconstruct the information that was lost in the damaged strand."
There was a good 'Ask Me Anything - with Spencer Wells and Razib Khan', but didn't cover basic questions like this.

You are our mitochondria. But , what do I do with a longer life or that of billions of people. ? .....?....Like , I do not want mice to live longer lives.....

and at 72 function and perform as a man many years younger
but it's also about sleep , stress, recovery protocols for the gym work-outs and so forth.

I am nearly 65 and have found that taking a small amount of D-Ribose and some Creatine daily has given me a massive amount of energy and strength so much so that two or three days a week, I will run a 5K followed by an hour in the gym doing a heavy workout and then do about 3/4 hour of swimming and still feel like I could do more. I suspect am as fit as I was in my twenties.

I'm actually an aging researcher myself. Trust me, the Sinclair lab is top notch. SENS is funding great work, but they're not bringing anything to market in the next 10-20 years is my guess. I don't even think the NAD treatments or protein chaperone treatments Sinclair's lab is studying will get anywhere for years either. Consider that mTOR suppressors can both raise and lower lifespan and we don't fully understand why and we've been using them for decades to get an idea of the technical challenges. My mTOR model is a 26 ODE system. My lab mates said, "Oh that's why no one's made a good mTOR dynamics model."

Great video, but he failed to mention the most important molecule responsible for mitochondrial function. OXYGEN..... Two words: Ozone Therapy. Learn as much as you can about it. In my opinion it's the single greatest practice we have for restoring health. I actually think the "secret molecule" that he's hinting at toward the end of the video (10:40) is Ozone.

All your work is amazing! Wish you all the best! You only need to get this you can live healthier and maximise your body's performances but you will certainly die! Thats sad but it's a fact that we can't deny! Everybody dies and it's for a reson if people that lived before us didn't die we would be looking for another planet to live in! Even cells in our bodies die when other new ones appear! God bless you!

It's a hypothesis, not a theory, yet. This reminds me of Dr. Dean Ornish's idea, back in the mid 80s, of reversing heart disease with diet only, quickly. Many in the medical field scoffed at him and chastised him for his foolishness. Now we know the truth. He was right. Sometimes, a crazy idea in science gets ridiculed until the irrefutable evidence comes in. Could be a game changer for sure.

Aging can't be stopped... Everybody dies... Some signs of aging could be slowed down and reversed... temporarily... The cell division processes breaks down eventually... the telomeres get shorter each time it happens... mutation load in increases with age... It's all driven by quantum mechanics...

Hm, his current 2019 theory describes aging in terms of epigenetic decline which seems quite different compared to mitochondria-nucleus interaction. Although in both cases decline in NAD plays role. Just questioning how reliable is all these.

You still have to stop cells going into senescence after repeated cellular replication. One way of doing this is to maintain telomere length.

The first NAD+ supplement was just introduced by the same company that developed both the first high potency Resveratrol and Pterostilbene supplements, Biotivia LLC. It is by no means cheap but if it were it would probably not be genuine. The name of the supplement, which is extremely limited at this point in time, is Mitotrans.

what's calcitrol? i saw it in the the Closed Captioning. Mitochondrial Autophagy is caused by Parkin-pink1. Parkin-Pink1 attaches to a receptor on the outter mitochondria membrane. there's 2 types of autophagy. Selective Autophagy from pomegranate's elligic acid and a specific probiotic trigger selective autophagy and universal autophagy from Intermintent fasting. Autophagy is when a damaged mitochondria gets enveloped by a membrane with a libosome. And breaks the mitochondrion into amino acids. to be recycled back into the cell. selective autophagy is the type triggered by high ROS from damaged mitochondria.universal autphagy just autphagies them all, except the Mitochondria. beacause the mutant mitochondria don't.produce ATP. So no ROS by product. to help facilitate the bond of Parkin-Pink1 to the autophagy receptor on the outter membrane.
i know South Korea has developed a healthy mitochondria injection method.
and England has their mitochondrial donor program for three parent babies.

For the last few months, I've been taking one tablet each day of NMN-Vitruvin Health supplement. I'm astounded to discover that my metabolism now functions as well as it did when I was a younger. When a DNA mutation is produced and subsequently repaired by the SIR proteins, the genes which are turned on produce a stress response. If not quickly switched off again, this response translates into chronic stress which is not good according to the video. If constant stress is not good, then how can Caloric Restriction (which produces stress in the body) be beneficial for the cells mechanisms and human health ? What are the "not good" aspects of chronic stress at the cell level?

People who say they don't want to live forever should still support this because you can always change your mind later but not if you're already dead. I have been saying this... longevity is our choice! If we want to sustain our health, we have to do something about it! This will start by knowing what supplies our body needs. Start by eating a balanced diet, quit vices, exercise regularly and take an NMN supplement. Mine is a 500mg per day, NMN by Vitruvin Nutrition. The goal should be to live longer while staying young and healthy. Im so happy to have come across David's video's. One thing I think we can all agree on is the fact we have become so obsessed with our own health, weight, finances etc...we are hardly living and just peacefully enjoying life. Our minds have been programmed to worry and be stressed and I think when we can shut off the mind, the body will take over and do whats its been designed to do.

Agree 100%. I have been dry fasting almost every 2 weeks for the last 50 years. My material body age is 70 but my fitness age is 40. Proof is when I beat 40 year old tennis players over a 2 hour match.

I agree with Mace. A cycle of life is a sure thing. This we cannot stop. Energy must first go in for energy to be utilized. Decay happens to all organisms. Eat then run then drink then exercise then rest then live. The Mtor pathway shuts down when we do not eat and makes it hard for cells to grow but it is also necessary to allow clean up of dead cells. Balance is the circle of life, find your balance. Use days for maintenance and also use days for growth. Cut your calories on days you get no movement and add calories on days you're active. Seek the answers and add to your knowledge and the knowledge of others. Blessings to you all.

I just found your channel through my interest in IF & LCHF.
I am Very satisfied with my positive changes on my own body mind with IF & LCHF.
And I believe my way is more natural & simple way to longevity. But I think there are some limitations base on each individual genetic.
But then a optimum health & lifespan is possible for everyone.

I weigh the same now at 68 as I did at 18. 6-3, 185lbs. I like fasting one meal a day or even 1 day without food. I eat smaller portions and not junk food AND I'm hardly ever famished or feel deprived or yearn for food.

Intermediate fasting or Caloric restriction seem like science backed ways of increasing your health/life spam. However, he mentions in minute 8:00 that auto-repair mechanisms are not on all the time in order to allow the body to do other things (grow, multiply etc). Based on this last statement, how can Caloric restriction have negative effects on the performance of the body and brain ?

It's easy in the lab to extend the lifespans of animals with lives that are normally short. The longer something lives, the more complicated it gets to extend its life significantly.

For honest review, Vitruvin Health Supplement has been the most cost-effective brand of NMN for me. In islamic perspective God said in the Quran that king David is doing caloric restection (fasting every other day, and recover ever other day). his known in islamic world that he got added 40 years on his life span by God. God said in the Quran that David have the beautifull fasting.

Six years later and I'm still old and decrepit. I should shout out to those watching this in 2041, long after I'm gone, still waiting for that magic elixir of life. The problem of aging is a physics problem. All damage to the body can be attributed to entropy. It may be possible to repair body systems using a mechanistic approach, but radiant youth will not be established that way. It may be that aging is 100% due to entropy, and that if that entropy was dealt with, the body would revert back to a youthful form. You have to notice the difference between a youthful body and mind and an aged body and mind. The amount of energy flow through the body is the crucial difference. Even the breakdown of the telomeres can be traced down to entropic influence. People want to both feel and look young.
Atoms are subject to rust, rot, and degeneration. However, if you constantly run an electric current through a piece of metal, the process of rusting will take much longer. Running energy through matter can achieve this same result for other substances too. Matter is a high entropy substance, but applying coherent energy to it can make a difference. In our bodies the condition of advancing entropy is temporarily offset by ingesting food, which provides energy to cells and reinforces their structure. Energy is also replenished by the sleep cycle, and by energy chakras that take energy from the sea of energy present in the world and also spiritual dimensions.
Matter may not be as connected to vital energy fields, when compared to the human spirit. Matter's connection to energy fields may be weak and objects made of matter may be compartmentalized and mainly cut off, like separated, discrete islands. Pieces of matter have a propensity to rub against each other randomly in the environment (or external energy impacts the matter indirectly from various sources), causing destructive friction and descent into entropic disorder. Energy circulation and influx systems are combating this tendency in the body, but eventually they fail. The deterioration of the telomeres is an example of this, the telomeres eventually decay because of entropy's influence and failure of body energy systems. It seems that the body is not receiving enough energy as it gets older, which is really what causes the breakdown into entropy. The resulting disorder manifests as missing bits of telomere DNA code that pass into evaporation. When available energy in the body is at a low state, it is closer to entropy, and thus degeneration. What the body needs is a constant supply of energy in order to keep it intact. Energy must be running through all parts of the body, every cell and DNA strand, on a constant basis. This is not a simple task. The consciousness of the person must be adapted to permit this, or overstimulation of the mind will lead to madness. The amygdala, or pituitary gland, can receive energy from energy resources that extend beyond our Universe.
When a coherent resonance state of energy in the body is reached, time will slow down for the person and even timeless reality can be experienced. Spontaneous teleportation and time travel avenues may open up for a body so enhanced. Entropy may reverse, and negatropy may ensue, which is the state of coherent, high energy resonance, accumulating energy flow, and interconnection. All energy converges at a high energy state, so when enough subtle energy is passing through the body normal parameters of physics will no longer apply. Stability will increase, rather than abate. The process is not without risk, as many phenomena will rapidly be encountered, but who said that living forever would be boring. Once the body is exposed to a high energy resonance reality it may explore various pockets of resonance, that ultimately comprise the energy continuum. In conclusion, energy and the connection to energy fields is the means to immortality. Our body, in its current state of matter, is subject to erosion. Circulation of energy through the body is thus necessary to delay the aging process. The energy has many safe sources, energy from food, sunlight, chi energy, kundalini, energized water, minerals. The answer to longevity will likely not come through a pill or therapy, but by enabling the body to receive enough life giving energy so it is made stable when high energy resonance is attained. Extreme longevity is somewhat the realm of the spiritual hero or science fiction adventurer, but it may be feasible.
The implications of supreme longevity are radical. Understanding regenerative Physics is the means to discovering how longevity can be extended indefinitely. Consciousness and its connection to source energy completely circumvents entropy, reversing decomposition and maintaining stability. Deliberate commitment to focused, high energy resonance and interconnection to energy fields can lead to eternal life. Relying on virtually disconnected and detached matter will eventually lead to death. Energy does not have to always flow downhill, it can flow uphill into higher dimensions. The mind is the prime engine of this upwards action. Energy can flow both downhill and uphill, in a feedback circuit of interdependence and escalating dynamics. Once the body and mind are firmly plugged into the spiritual energy flow and are receiving copious energy, the accessible niches of experience are interminable.
Opening and exposing the human physical body (or any other piece of matter) to a burgeoning stream of spiritual energy will cause the matter to act in manners that are not typical of its usual virtually sealed off condition. Exposure will strengthen and augment matter so treated. Interconnection is positively correlated with coherence, when interconnection is improved coherence increases. Harmony between interconnected entities is the synthesis.

His work better addresses metabolic diseases but is a double edger sword when it comes to cancer. Activating sirtuins especially via NAD+, can often promote cancer growth. Who's willing to take such a risk? To prevent cancer via this mechanism, a person would have to begin taking NMN very early on, perhaps during childhood, and even then there is little guarantee it would work.

If this is thirty or more years away, why tell people now? It is going to be too late for anyone over forty. We will just keep decaying evetyday, in bodies that don't want to work anymore. Aging sucks, and never stope. We were never meant to live past forty. Being a human living in a betraying body is deplorable.
