NMN, NR, Resveratrol, Metformin & Other Longevity Molecules | Lifespan with Dr. David Sinclair #4

NMN, NR, Resveratrol, Metformin & Other Longevity Molecules | Lifespan with Dr. David Sinclair #4 302,782 viewsJan 26, 2022 Dr. David Sinclair and co-host Matthew LaPlante zero in on drugs and supplements that have been reported to combat aspects of aging. They share the latest experimental and clinical data for NAD boosters (these being NR, NMN, NAD IV drips and shots), resveratrol, fisetin, quercetin, rapamycin, spermidine, metformin, and berberine. Given the abundance of data available, a special focus is placed on the NAD precursors nicotinamide riboside and nicotinamide mononucleotide. Known mechanisms, limitations, and/or side effects associated with these molecules are additionally highlighted. Correction and clarification: At time point 1:07:37, Dr. Sinclair says he takes 1 gram of Spermidine, but the active ingredient in the capsules is 1-2 milligrams. We apologize for the confusion. #DavidSinclair #Longevity #Lifespan Thank you to our sponsors: Timestamps: 00:00:00 Kicking Off Episode Four: Longevity Molecules 00:02:15 Thanking the Sponsors 00:04:46 An Additional Boost Beyond Adversity Mimetics 00:05:52 Drugs vs. Supplements and Highlight Points 00:09:54 Longevity Molecules Target the Survival Circuit 00:11:33 NAD Boosters 00:13:32 Nicotinamide Riboside 00:21:26 Nicotinamide Mononucleotide 00:29:22 NAD Intravenous Drips 00:32:45 Bioavailability of NAD Boosters 00:33:40 NAD and Cancer 00:35:33 Resveratrol 00:42:26 Red Wine and The French Paradox 00:43:08 Fisetin and Quercetin 00:47:09 Rapamycin and Rapalogs 00:49:10 Spermidine 00:51:27 Metformin 01:00:27 Berberine 01:02:43 Dr. Sinclair’s Protocol for Longevity Molecules 01:08:51 Wrap-up and Next Week’s Episode: Interventions on the Cutting Edge 01:09:52 Options for Subscription and Support Links: The Sinclair Lab website: https://sinclair.hms.harvard.edu/ Nicotinamide riboside is well-tolerated in humans: https://go.nature.com/3G1blbb Nicotinamide riboside is a sirtuin-activator: https://bit.ly/3FUWWxa Mice treated with nicotinamide riboside are longer-lived: https://bit.ly/3KD5L2h Nicotinamide riboside protects mice against obesity: https://bit.ly/345E4hF An inflammatory signature is attenuated by nicotinamide riboside: https://bit.ly/3Aum78I Nicotinamide riboside, pterostilbene, and ALS: https://bit.ly/3nYdxK6 Nicotinamide mononucleotide combats diabetes in mice: https://bit.ly/3AwakXx Aging is slowed by nicotinamide mononucleotide: https://bit.ly/344jGgW Nicotinamide mononucleotide in prediabetic women: https://bit.ly/3H1vv6b NAD and cancer in mice: https://go.nature.com/32xl1wp Resveratrol protects mice against a high-calorie diet: https://go.nature.com/3G2pZyE Resveratrol in patients with type 2 diabetes: https://rsc.li/3Aupk8h Supplementing resveratrol in metabolic syndrome patients: https://bit.ly/3IE6C0Q Fisetin enhances longevity in mice: https://bit.ly/3o10hom Quercetin and fatty liver disease: https://bit.ly/3H4KWdF Dasatinib and quercetin decrease senescent cell burden: https://bit.ly/35pCHuY Mice fed rapamycin late in life live longer: https://go.nature.com/3F0yud3 Spermidine induces life extension and cardioprotection in mice: https://go.nature.com/3fXL5nn Spermidine and older subjects with cognitive decline: https://bit.ly/3IG0MMy Healthspan and lifespan in mice are improved by metformin: https://go.nature.com/3JSRZbs Metformin, age-related diseases, and mortality: https://bit.ly/3KG0iaS Targeting aging with metformin (TAME) trial: https://bit.ly/3eU9ABk Treating mice with berberine extends lifespan: https://bit.ly/3AwkO9l Lifespan in fruit flies is prolonged by berberine: https://bit.ly/3nYpwYt Berberine and metabolic disorders: https://bit.ly/3KUpwCC Please note that Lifespan with Dr. David Sinclair is distinct from Dr. Sinclair's teaching and research roles at Harvard Medical School. He has a very effective way to be both a “layman” as he asks questions while at the same time, generally savvy on the topics. Also, I congratulate you both on not taking yourselves too seriously. We already get enough of that with certain scientists. 299 ashley bryan ashley bryan 2 months ago (edited) I was so stoked when I saw you started this channel. Loved watching you as a guest on other podcasts, so this is a treat. Great job so far! 162 Nicholas Arapis Nicholas Arapis 2 months ago (edited) I aged a full week waiting for this podcast 🙃 498 Red Pill Lense Red Pill Lense 2 months ago Thank you, Dr. Sinclair! I look forward so much to this podcast series. You truly are helping me 🙂 45 DoNotAge Team DoNotAge Team 2 months ago Another excellent podcast David - we love your work! 59 Tanner Revak Tanner Revak 1 month ago Great discussion! Thank you. I'd like to know if there is a correlation between stressed vegetables having greater health benefits VS a stressed animal. I can imagine there is a whole different set of rules for an animal that has been penned up and fed a terrible diet, VS a plant being harassed by bugs, weather, fungi, virus, etc... Keep up the great work! 7 Marwa Mite Marwa Mite 2 months ago Hi! Thanks for all your work - When you were on Huberman's podcast, you briefly mentioned that taking Resveratrol on alternating days produced better results in mice based on a specific study, and that the beneficial effects could even be mitigated by daily intake. But I've also heard you talk about it being in your daily stack, as you've mentioned here. Could you elaborate/clarify whether you think a daily or alternating intake could produce a significant difference in potential effectiveness of Resveratrol ? 82 M M 2 months ago Dr. Sinclair, hope you will discuss how NMN affects kidneys. Kidneys are crucial to life but are not discussed often. Thank you! 33 Spirt Fire Spirt Fire 2 months ago What are your thoughts on apigenin? The shows notes, the timestamps and the explanations were very digestible (pardon the pun). Seriously thank you David, Matthew, And the team at Harvard medical school working hard to bring us the best longevity science to date. I can't believe that this information is freely available for all of us 2 Jen Pal Jen Pal 2 months ago Thanks for this podcast, I appreciate all your work and I been trying to follow your regimen. I wish that you could tell or advise us which you think is the best choice NMN supplements. I been paying a lot for it and still not sure if they are true to their advertising or I pick the right one. 14 Rosie Smallshaw Rosie Smallshaw 2 months ago Love your podcasts! The synergy is always courteous and encouraging. Keep up the great work 👏🏼👏🏼👏🏼 5 Paul WN Paul WN 1 month ago Really loving the podcast series. Great refresher for the book. Finding some of the supplements in Australia is harder than the US. Any recommended labs? 2 Boyd Hooper Boyd Hooper 2 months ago Great that you go deep into dosage, timings and other details on the supplements. So frustrating when others don’t do that. 9 Thomas B Thomas B 2 months ago I was hoping that you would discuss Methylation concerns and the use of TMG to mitigate. Also, I find the piece that’s often missing from Such discussions is for pregnant & breastfeeding mothers. 30 Suncloud Mashups Suncloud Mashups 2 months ago Thank you for another great episode. I'm really a fan of these. I have a question about NAD boosters - do you think that supplementing with something like NMN could potentially suppress the bodies natural production of NAD in the long or short term? 32 Levon Darratt Levon Darratt 2 months ago They are both great. And co-host - just the way he breaks things down - while being super educated as well. And stops things, when they get confusing. Then David re-iterates, and explains as needed. Those 2 are something else! 4 Rapamycin News Rapamycin News 2 months ago Great to see David covering rapamycin a bit more than in the past. 6 That Chess Guy That Chess Guy 2 months ago Hi David! I’ve been implementing what you suggest (I haven’t yet implemented supplements yet) and I feel a lot better! One of the things I’ve done that’s helped a lot is actually something you haven’t mentioned in the podcast yet. I took a food sensitivity test and cut out the foods that tested positive for sensitivities. Milk was apparently causing me muscle and joint inflammation as well as stomach problems. 29 Futons Japan Futons Japan 2 months ago Great stuff. Hopefully we can get a list of blood test to take and how often to measure effectiveness and if it causes any problems. Apart from the physical or mental impact 2 Mick Wannink Mick Wannink 2 months ago Thanks for all the info! I would be very interested in the question from which age it makes sense to use things like NMN or NR. With it comes the question to determine the levels of NAD currently available in the body. Is there any quick and easy way to determine in order to avoid spending money on supplementing in case the levels are good? Or is there a general benefit for everyone to take it, like e.g. for vit D3 which is generally low in the population of countries that dont see too much sunlight? 18 R. David Young R. David Young 2 months ago (edited) Thank you! I appreciate your information and the sense of respect you seem to have for human kind and life in general. I wish both of you vibrant health and long life. 💚 I listened to your book on Audible. It's great. 3 Puppygirl FM14 Puppygirl FM14 2 months ago David, thank you for sharing your knowledge to help others! Question about Berberine- when should I take it? At night, the same as when you take Metformin? 6 Mrs. Marie Mrs. Marie 2 months ago How much berberine should you take to equal Metformin? I would like to take NMN but am now worried about it causing cancer growth. Great podcast! I am glad I found it! 12 Katharine of Arrogant Katharine of Arrogant 2 months ago (edited) Highlight of my week. As soon as I listened to these I started OMAD, changed my diet and exercise routine. I've sent this to all my friends and family. Pure gold! 13 Tom Wawersich Tom Wawersich 2 months ago Love the podcast- great episode! I have heard I should be taking TMG with NMN - would love to hear if you do this. Chenoa Siciliano Chenoa Siciliano 2 months ago First of all thank you for this very informative podcast. It would be so great as a follow up to this if you could include information on: Methylation and TMG with MNM; Cycling molecules like Resveratrol, Fisetin, Quercetin. Also I am very interested in Cycling Berberine, for it to be more effective but also because I have read and stand to be corrected, but from what I have read it is antimicrobial and you can risk wiping out your gut microbiome, especially if you are taking a gram a day! And then lasty are you still taking aspirin daily, If so at what time of the day are you taking it, is it after food to protect your stomach, and is t daily too are do you also cycle it? Oh one more thing a lot of molecules are hormonic stressors can you do to much and risk actually becoming too stressed. like if you took molecules that mimic fasting, and you fast and you exercise and you have a good amount of normal daily stress that you try to keep under wraps, do you risk a kind of burnout? Thanks again! 8 Mayra Kennedy Mayra Kennedy 2 months ago amazing episode! loaded with so much information. question about a supplement called,True Niagen. I took it for about 3 months but didn't notice any differences in my skin or performance. What time frame do you recommend taking a supplement for anti-aging? 6 month? a year? Thank you :) 1 Tyler Eidt Tyler Eidt 2 months ago Thanks for this informative video! Couple questions. If resveratrol works better when ingested with fats, would it be as effective to take with fish oil capsules as with the suggested yogurt or olive oil? Also, would taking Athletic Greens in the morning significantly break the fast if trying to skip breakfast? Thanks again! 2 MJ MJ 2 months ago I discovered Dr. Sinclair from Dr. Andrew Huberman's youtube channel. This is amazing information from Dr. Sinclair. Thank you for sharing your knowledge! 3 Win Balingit Win Balingit 2 months ago I saw Dr. Sinclair on a podcast giving a lecture on Resveratrol, it was for a wine convention, everyone, including Dr. Sinclair was drinking red wine!! The lecture was very educational and entertaining!!🤩👍🏽 7 busysuperhuman - Sara Pugh busysuperhuman - Sara Pugh 2 months ago Thanks for this - I love the way you and Matt interact - All scientists need a Matt to help explain to lay people 2 mrLoftladder mrLoftladder 2 months ago I'm taking a similar supplement regime myself and have been doing it for a couple of years now. 67 Y.O and feel more energetic and certainly fitter than any time since my early 50s. loving these podcasts learning so much from them, thank you. 3 Cheryl R Leigh Cheryl R Leigh 2 months ago Great podcast series; very much appreciated. 2 Gunnar Gunnarsson Gunnar Gunnarsson 2 months ago This is exceptionally interesting. I wonder if there are any studies addressing the additivity of the benefits of different supplements! If two different supplements increase the lifespan of mice by say 30% each, what is the combined effect of the two? 7 Francis Chen Francis Chen 2 months ago Thank you David - I am a huge fan of your work! 2 Phil C Phil C 2 months ago Id been waiting for this one . Thank you so much for these podcasts, keep them coming . 1 Guillermo Torres Guillermo Torres 2 months ago Thanks for the podcast! My request for the future is to do a deeper dive on some of the markers. I’ve been very frustrated in particular about VO2MAX. But would be good to understand the ones from Inside Tracker as well. 1 A Fath A Fath 2 months ago Thank you for this podcast! You were taking baby aspirin at one point to prevent bowel cancer, have you removed that from your protocol? I was diagnosed and cured of bowel cancer but I am not sure if I should start taking aspirin daily for prevention 4 Felix R. Felix R. 2 months ago Thank you for another very informative episode! I have a question though. At what age do all those measures even start to pay off? I'm 26 years old and I live healthy. Everytime you mention a supplement I ask myself things like "Do I even have senescent cells, that I need to kill? At what point in the future should I start taking quercetin/NMN etc. ?" 3 Waldiney Torres Waldiney Torres 2 months ago I'd love to know Dr. Sinclair's opinion on stereoisomers of resveratrol and pterostilbene, wheter it is just marketing or something else entirely. Fascinating conversation as always. 3 Lucy Cooper Lucy Cooper 2 months ago Just finished his book . I was glued the entire time. I know he’s working on another. Anyone know it’s approximate release? I can’t wait 🏋️‍♂️ 2 Kimbo slice Kimbo slice 2 months ago Grateful for the information you guys are offering 🙏 3 John Wilhelm John Wilhelm 2 months ago Have been following David since the early Resveratrol days. Was super excited when this series started. Thank you for your continued efforts to expand our knowledge on this topic. You and Matt make a great duo and I hope this series continues beyond the current scope. 4 Joseph Benjamin Joseph Benjamin 2 months ago Love Your Work Professor. Thank you for all your hard work and willing to share your findings with us. I live in Sydney Australia. unfortunately, as you have mentioned many times none of these would matter unless we frequentley mesure and see how these molecules are effecting us. I am fining it deficullt to find clinics here in AU for instance to provide Biological age. Any chance you can provide some guidence on how we should go about it? 3 Ken Ken 2 months ago After starting NMN,Resveratrol and Berberine with central and obstructive sleep apnea my deep sleep has improved by leaps and bounds. 3 Captain Pike Captain Pike 2 months ago Important to emphasize ‘the threshold level’ as David discussed in his interview with Rhonda Patrick PhD , the ‘natural clearing process ‘ of the body that will eliminate NMN supplements , Resveratrol from the body - and David’s idea of ‘pulsing ‘ these supplements every other day , then fasting and exercising 7 Anna Bilous Anna Bilous 2 months ago Thank you for this podcast, very interesting! Sorry, but for non-scientists could you please explain whether taking NADH provides an NAD boost as well (the one I am referring to has Niacin NE (750mg) & NADH (6mg))? I read that NAD/NADH ratio is important and higher NADH would seem to speed up ageing... That doesn't mean that the NADH I am referring to (that is mainly Niacin NE) is not so good for ageing, does it? Sorry if this is a silly question! 2 Pavel Drobnych Pavel Drobnych 2 months ago (edited) In recent studies they show that resveratrol/pterostilbene induce replicative stress - should we get concerned? Otherwise this is a truly super-star show, many thanks for doing this. gg84 gg84 2 months ago Thanks for the very informative podcast and also talking about studies which show harmful effects of some of the supplements. For folks outside US, where we don't have inside tracker, can you pls suggest periodic blood tests we can do to ensure that the supplements aren't having a harmful effect on our bodies (including nmn potentially expediting cancer cell growth) ? It will be great information and people elsewhere can keep a tab on their health with these supplements. 32
