Mammalian target of rapamycin (mTOR) was named because a drug called rapamycin inhibited a protein that was then named, target of rapamycin (TOR). The “m” is because it’s in mammals (zebra fish TOR is, similarly, zTOR). mTOR is central to the opposing biological processes of growth and longevity. mTOR is thought to promote growth and decrease longevity. Basically, any process you can think of relating to muscular growth and longevity probably has some relationship to mTOR. Here are some examples: Exercise activates mTOR signaling in skeletal muscles (growth) mTOR inhibits autophagy (longevity) Insulin activates mTOR (growth) Sirtuin proteins inhibit mTOR (longevity) BCAAs activate mTOR (growth) Fasting inhibits mTOR (anti-growth and longevity) And, as precited by its pathetic name, rapamycin increases (longevity), at least in mice How exactly does mTOR impact all these processes? Well, it’s kind complicated [show image below] and there are actually two different mTOR complexes (mTOR1 and mTOR2) that have slightly different functions. So, for the sake of simplicity let’s just say this: mTOR is a kinase. This means that mTOR acts to regulate the activities of other proteins by adding a phosphate group from ATP to them. Depending on what protein it adds the phosphate to, and where on that protein it does so, is can either activate of inhibit its target and pathways downstream. The message, in its simplest form, is that mTOR activity makes you live hot (and strong) but die young. But since when is science ever simple? What annoys me is that people tend to be scared of anything that activates mTOR because they are afraid it’s going to kill them. This is problematic thinking for two reasons: (1) Biology is all about balanced cycles. You need periods of growth (anabolism) and breakdown (catabolism). If you’re all anabolism (like a body builder) you will die young. However, if you’re all catabolism, you’ll die even younger. In fact, one of the leading risk factors for death in the elderly is low muscle mass. You need a functional level of muscular power to live and thrive. No mTOR and you’d wither away. In short, you do want to activate mTOR on occasion. Just don’t overdo it. “Extremes are easy, the challenge is balance.” “ (2) The inside of your body is NOT a homogenous blob! Why do people always forget that fact? Just because you activate mTOR in once place doesn’t mean you activate it everywhere. For example, when you work out you activate mTOR in your skeletal muscle but might actually decrease mTOR activity in your brain. Thus, it’s possible, and I would argue probable (such studies can’t be done in humans without cutting into their brains), that exercise activates muscular mTOR but inhibits brain mTOR, giving you the exact organ-specific balance of activity that you want! I have been looking into the relationship between mTOR and longevity (or the lack thereof). New research suggests over activation of mTOR pathways may be a key area in shorter life span. If you make another video on this subject, I will watch! 9 Reply @sandraat @sandraat 3 years ago (edited) Yes please more on the scientific part of mTOR. I think you are doing a great job explaining the science on your subjects. Please keep it coming!🤩🙏🏾 11 Reply @1yoan3 @1yoan3 3 years ago (edited) I love your videos, and the more nerdy and detailed they are, the more I'm fascinated by them. I hate just random facts thrown around like a lot of youtubers do, Thomas goes into great detail and I love that, he explains everything. 6 Reply @rmohamed786 @rmohamed786 3 years ago (edited) Wonderful explanation on how to benefit between ‘Mtor on’ and ‘Autophagy on’ cycles. I started the Ramadan fasting of 30 days where one goes for about 14 hours of dry fasting everyday. I am going to alternate between foods that promote autophagy when beginning the fast before sunrise and foods that promote Mtor activity when breaking the fast at sunset with some exercise just before that. I think this will be a great strategy to balance these two states on a daily basis. 4 Reply @olgap.2902 @olgap.2902 1 month ago Firstly, love your channel and thank you for bringing all that knowledge to us, and in a clear to understand language. Would love a more detailed video since mTOR is something I'm actively conscious of and think about (I do resistance training and alternate fasting). Anyhow, I thought mTOR gets turned on when consuming at least 2g of leucine (and with exercise you just stimulate protein synthesis and that's all, so a new thing learned here). I even count my leucine intake when, for instance, I don't want mTOR activated (but consume at least 30g of protein with each meal). So after seeing this video, I admit I am a bit lost with this mTOR thing ... Thank you in advance, and once again hugely appreciate your channel! Reply @opedromagico @opedromagico 1 year ago (edited) I love how simple you explain stuff! Thanks, Thomas! Keto carnivore here for 1 year, it saved my life. I'm a layman (no training in health sciences) but I'm really interested in this. I'm especially interested in practical recommendations of how to build a routine that achieves the balance you're referring to. SiimLand has some videos offering sample workout/diet routines designed to achieve this but I'm looking for more sources. This video was helpful and a good primer on the topic, thanks Thomas. Reply @wszystkozajetejuz2 @wszystkozajetejuz2 3 years ago (edited) very interested in mtor and balancing muscle growth with longevity. the question i'm asking is should i maintain high protein intake all the time or should i cycle it. I'm especially interested in cycling of autophagy days/weeks (e.g. fasting, metformin) and body building days/weeks (+ high protein, bcaa intake). 13 Reply 1 reply @La_sagne @La_sagne 9 months ago finally an explanation. ive heard so much about mtor is good for muscles and strength is great for longevity because falling badly when youre over 65 has quite a high lethality rate and then i heard rapamycin inhibits mtor thats why its great for longevity and i was very confused. it sounds a lot like cortison the way you explain it. cortison is important in stressful situations like a workout and afterwards for recovery but you dont want chronically high cortison levels because that has a bunch of side effects that kill a lot of people in their 50s or 60s already 1 Reply @elliottclark8776 @elliottclark8776 1 year ago (edited) Wouldn't mind extra info on how MTOR is activated and works within the body. Reply @vincentperry6791 @vincentperry6791 3 years ago (edited) Hell yea, this is exactly the stuff i wanna do more research on... been self expirimenting through keto IF for the past several years and i can say from a medatative perspective you can definatly feel the difference in consciousness between the two states and which one your bodys leaning more tword... what I'm super intrested in finding out is the ratio between the two to acheive ones optimal balence over the function of time... like you said its not a switch and, both processes are usually happening simultaneously one more so than the other but hypothetically if from a balanced state i spent 24hours in 100% mtor building mode, then would 24hours of 100% autophagy recycle mode restore the balance? I suppose this is an impossible question as everything is individualized and situational but don't you think it'd be cool to have some sort of device, kinda like hoe a glucometer works as to tell you which state youre in and how much so? I do have a question if you don't mind in regards to Intermittent Fasting......Technically speaking when can your time start after you have finished eating your last bite of the day? In others words can your fasting window start immediately after your last meal or should it start like 30 minutes later an hour later? Stupid question I know, but I'm sort of OCD and anal when it come to exact time windows for starting and ending fasting periods. Thank you for your help. Wainting for Thrive Market to start operating in Brazil 🙄 Reply @hemangpatel6239 @hemangpatel6239 2 years ago (edited) Thank you for making a video on mtor. So considering mtor is activated during weight lifting, should I always consider breaking a fast right after my workout instead of breaking a fast 6 hours later so that I can reduce the number of times mtor is being activated throughout the day? I prefer morning fasted workout and not eat anything after that and then break fast at lunch and have a dinner and so on. As I expressed my concern above, I want to reduce the number of times mtor is being activated throughout the day but at the same time want to gain more benefits by combining a better routine. Also if you can clarify if I really have to have a protein shake or meal after a workout opposed to going faster for 5-6 hours after working out. I'm a big fan of your research and clarity of the content you provide. Thank you for everything. If you want to slow the aging process, then you should be taking a look at balancing the activation of the mTOR pathway with the AMPK pathway. Both are needed as an aging adult, yet are mutually exclusive. The mTOR pathway is great for growing new muscle and staving off sarcopenia, but the AMPK pathway is required to clear out the toxic by-products that mTOR create. In this video, we’ll be talking about what the mTOR pathway is, and how too much of a good thing is not good for you. We’ll also go into how balancing the mTOR pathway by activating AMPK is a good thing. And finally, we discuss how to do both, and achieve optimal balance between the two. Nice new format, the CC is better positioned on my screen. Excellent use of infographics. Fun puzzle of what's the best mix of HIIT, weight training, amino acid supplementation and, in my case, my weekly dosing of Rapamycin? I'll be researching weight training recovery times, maybe doing the most resistance training 3 days after (and 4 days before) my dose of Rapamycin. Since I've been on Rapamycin (5 months) and increased my HIIT training, my resting heart rate is lower than it has ever been in my life - including my extremely athletic state at age 30. My athletic performance now is pretty good and I am tracking it. Please continue the science detail in at least some of your videos, perhaps researchable links, as well. 1 Reply Lance Hitchings · 2 replies @stanleystark1842 @stanleystark1842 9 months ago Love your explanations. In one of Dr. David Sinclair's talks he mentions Metformin and Green Coffee Extract both activate the AMPK pathway. Presumably alternating between resistance training/essential amino acid days (anabolic days) and Metformin or Green Coffee Extract and less amino acid days (catabolic/autophagy) with rest would be one method of pulsed therapy. Dr. Sinclair emphasizes that no one knows yet what the optimal regimen is for this kind of anti-aging therapy. It would be great if people pursuing it would keep records and do either the DNA methylation blood testing or telemerase length testing and contribute their data to some centralized database so that we could determine what the optimal protocol is. I believe William Faloon's (of The Life Extension Foundation) non profit organization is compiling such a database but I forget the name of his non-profit at the moment. I don't really lift but I hit the Bowflex a couple times a week and I like to follow that with a big mug of hot chicken sipping bone broth into which I have added 20 gm of collagen peptides from cow hide. It has all the AA's except for tryptophan and best of all only 106 mg of Methionine. At 90 kg body weight my RDA for Methionine would be about 900 mg and this leaves me room for the egg, goat cheese, and fish in my diet. This seems to work pretty well for me. Liked the video. Kinase is KY naze What you propose is consistent with what David Sinclair of Harvard, a major anti-ageing researcher (I’m sure you’ve heard of him) and others seem to be saying. I think this is a very complex subject and while I am following a similar regime I think the current picture will develop with time. Current thinking may then appear slightly naive. But that’s inevitable of course - humans can’t be researched like lab rats/mice. Presumably, autophagy cranks up because there then is a lack nutrients. Thusly, mTOR would not be activated. Then, when I'm not fasting and doing HIIT, mTOR would be inhibited boosting AMPK allowing autophagy too. Could you comment here s'il vous plait? Your muscle growth and repair doesn't just take place on the days you weight train, certainly not only 2 hours after when you take your amino drink. You do your most growth and repair on the days you rest. Also, while HIIT training on your non weight training days activates AMPK instead of MTor, you are still damaging muscle tissue and you need protein for repair as well. But I’m in psych and I was curious about something. Increasing MTOR signaling stimulates neuritogenesis, this is how psychedelics grow ur brain, how do we activate mTOR neuritogenesis while increasing longevity. Can we inhibit mTOR for most of the day but then stimulate it when we are in that really hard class? What are the durations of mTOR activation? Prussian Blue is different, but comes to mind because at one time, I was the principal investigator for Prussian Blue (Radiogardase), an orphan drug, which is used for both Thallium and Radiocesium poisoning. I slept well but when I woke up I felt very in control of getting up at any time I wished. I normally set my alarm on snooze but didnt feel like I needed to as I was tired but somehow alert. I had a UTI and it was a Sunday morning so I couldn’t find a doctor in the small town I was visiting before I had to catch a flight back next morning. It definitely resolved the symptoms - probably faster than some antibiotics. It didn’t clear the infection but it gave me a chance to make it back home comfortably and then arrange to see my doctor a few days later for antibiotics. That being said, I didn’t notice a remarkable improvement in energy but, between the uti and jet lag, I wasn’t feeling nearly as crappy as one would expect… 🧐 35 Reply 6 replies @iulianaa2079 @iulianaa2079 3 months ago In our neck of the woods, when I was a small child - 40 years ago, Eastern Europe - the doctors used it against throat infections. 14 Reply @genesmith3582 @genesmith3582 3 months ago nice topic. But yea, the reason Methylene Blue acts like an antidepressant completely unlike other antioxidants which are for the most part sedative is because at initial high concentrations it's actually pro-oxidative. Quite powerfully so. It'll dry out the skin around your mouth after you apply a ml to the back of your throat (common sore throat treatment). So initially it prompts the body to produce more of its own antioxidants... then becomes a net antioxidant when it eventually dilutes. It's so powerful that it used to be used as a treatment for cyanide poisoning. 2 Reply @Whatisthisstupidfinghandle @Whatisthisstupidfinghandle 3 months ago (edited) 7:30 When ROS interacts with fatty deposits in your skin it rusts them.
