I read The Sirt Food Diet book (Goggins and Matten) a few years ago and now incorporate a lot of the foods in my everyday diet. Is there a way to measure the levels of all the Sirtuins in our bodies (and also levels of NAD)? Fucoidan is not the name of a specific chemical substance, but rather the name for a class of substances. There are several different types of fucoidan, each with different molecular structure and different properties. Fucoidan refers to a type of polysaccharide that contains substantial percentages of L-fucose and sulfate ester group, but can also contain other monsaccharides, including mannose, galactose, glucose, xylose, etc. plus other compounds like uronic acids, acetyl groups and proteins. What type of fuciodan a compound is can depend on the type of seaweed that is was extracted from and the method of extraction, to name a few. In light of this, it's not too difficult to see why it can be so hard to find fucoidan that can activate SIRT6. I think your decision to create a longevity course and charge for it in order to support your work on YouTube is brilliant. Your current videos bring all the detail of the complex chemistry and biology of longevity down to a level of understanding better than anyone else on YouTube. I know it takes a lot of time and work to do that, and I’m glad to support you in your ongoing efforts. This will, no doubt, be a win-win for everyone. You get support and your audience gets a pre-digested source of understanding and information for the ongoing longevity research and the resulting practical capabilities being produced in this complex field. Here are two aspects of the course that I personally would like to see. First, I would really like to have the course content available in some “printed” form in addition to or instead of video. The amount of information involved and its complexity are very large, and just having a video to watch in order for me to mentally capture everything is not adequate. If only video content were presented, I’d have to spend a large amount of time taking notes and creating complex diagrams that I could use for study and review to ensure that I can confidently reason about what I should be doing to extend my own longevity. Ideally, the course would be presented both in video and in a printed format, such as PDF. If that were done, we students could view the video first to get the big ideas involved, and then study the text and diagrams to cement the details in our heads. Second, a hybrid of one-time purchase and a subscription model of pricing might work well. A person could buy the course to get the first version. The course would have to be updated occasionally, maybe on a quarterly basis, in order to keep up with the rapidly evolving field. So a person could either re-buy the course at full price whenever they think it’s worth doing, or they could subscribe to updates after they have purchased the full course so they could have lower cost access to the updates on a continuous basis. And if you really want to confuse everyone, you could also create two tiers of the course – one lower cost tier of video-only, and a higher cost tier of video and printed material. I’m not trying to tell you how to run your course, Lance; I’m just casting my vote for course features I’d like to see. However you do it, I’m sure it will be a major contribution to the many people who don’t have PhDs in this field but still want to understand it in order to extend their lifespan and healthspan. When the people making it and selling it are the same ones testing it there's a potential for fraud. Got terrible stomach cramps that needed to treated with an infusion. They asked me what I had taken, I told them about the product. They never heard of it - of course. I am sure the cramps came.from the capsules. I never had pain like that before in my whole life. I won't take any of these capsules ever again. Max lifespan is how long the oldest mouse lives. If most mice live 48 months, then 30% increase is (48 x .3). If Max is 58 months then max is 58 x it's percentage increase. One of the things I discovered, which was brought to my attention both by the geneticists and my doctor, is that I inherited a very rare version of SIRT-6, where the protein kinase is catalytically overexpressed by several fold. The guy from the lab send me a "Congratulation!" card, because, according to him, that SNP was found in two supercentenarians, which is not a small number since supercentenarians, especially male ones,, are exceptionally rare. Then, I thought about my family and I realized that it might be true. My paternal great-grandfather smoked 2 packs of cigarette a day for 50 years, and he died at the age of 109, just 4 months shy of 110. His brother died at 98...from a fall. My grand-uncle is still alive at 103, still walks without a cane and his only complain is mild arthritis to his joints. My sister also inherited the SNP, but unfortunately for her it is not properly expressed in females. Then, I thought about myself and I realized that there are patterns. I am 41, and people consistently tell me that I look like I am in my early twenties. I have zero grey hair either on my head, face or body. And just a couple tiny wrinkles around my eyes when I smile. My blood sugar never goes above the low 90's, even if I eat ice cream. On the other hand, when I fast, my blood sugar is higher than that of a fasting normal person, but still below that of well-fed normal person. I've never measuired my IGF-1, but I have measured my GH level, and it is always at the bottom 20% of people. I even considered supplementing with GH because of this, but then realized that whatever benefit from GH I would just increase my cancer risk. It was always kinda weird to me that my blood sugar never dropped much when I fasted, and I would get thirsty as hell. But the primary effect of SIRT-6 is not metabolic, related to IGF-1 regulation. The primary effect is increased DNA repair, especially of double-strand breaks, the kind that causes cancer. That is probably why my great-grandpa smoked 2 packs a day for 50 years but never got cancer(maybe). He also loved hard distilled liquor, and never got liver or throat cancer either. SIRT-6 also upregulates telomerase to lengthen short telomeres, which probably helps with things like osteoporosis and sarcopenia, since it helps to maintain stem cells. But not everything are roses for me. The same genetic testing revealed that I did not inherit the protective SNP of FOXO3A, which is something I expected since I am over 6'3 tall. I also have the MET-MET version of the COMT gene, which probably explains why I am so anxious and agitated. In fact, my anxiety is so bad that I will probably not live very long. Stress is one of the things that most shortens life. So even though I inherited a "super" longevity gene that allows you to live over 110 years, I might actually die at 80 or 85 from being such a worry wart. Point is, we are all kinda mixed bags. Some blessed in some ways, some in other ways. My MET-MET COMT allele. for instance, allows me to concentrate, but makes me a nervous wreck. As a big guy, I also don't have the protective FOXO3A allele, and those alleles are the most consistent findings among centenarians. Maybe my ST version of the FOXO gene will carry more weight in determining how long I will live than my protective SIRT-6 allele. Who knows? Too little quercetin and SIRT6 is down regulated, and then positive outcomes from some herbs used as inhibitors of SIRT6. Looking into it here for a few minutes I am not sure what is the most cost-effective way to incorporate it. We have full spectrum brown seaweed, extract standardized to fucoidan, and extract standardized to fucoxanthin, maybe other options. I've stopped taking resveratrol for now, but still take 300 mg NR daily. PS: Tried hyaluronic acid again; had same problem as before - vertigo apparently caused by swelling of inner ear. HA appears to do a good job of increasing internal fluid levels, but I don't seem to have the space for it in my inner ear cavity. I am somewhat new to my own efforts and research for longevity, I am currently 60 years old, started taking resveratrol about 3 years ago, then recently started taking NMN about 3 months ago, but recently stopped after you shared your research and I changed to take NR supplement instead of NMN, and I still take a pure form of resveratrol each day. I am very curios about DoNOtAge SIRT6 activator and I considering trying it, I am wondering though should I stop taking resveratrol given that studies have shown that increase in SIRT 1 along with increase in SIRT6 in mice did not have any longevity affect, only the SIRT 6 boost increased life span. So it seems that it would be a waste of effort and money to try to boost both SIRT 1 and SIRT 6 at the same time. Any feed back about that is appreciated. Anybody have any experiance with DoNotAge SIRT6 activator? I'm 55 now so it's going to be a while before I know if it's had any effect. Lis Parrish used gene therapy to start the gene responsible for telomerase production that is showing promise. SIRT6 is a deacetylase enzyme on histones, modulating gene expression. SIRT6 is also a mono-ADP-ribosyltransfarase and may restore adequate homologous recombination lost with aging. However, prostate and breast cancer cells express SIRT6, and it seems to confer increased proliferation to those cancer cells. Thus, mire studies are needed to clarify this issue. Also the only company that processes the algae and sells the Fucoidan only allows their own labs to do the testing. And oleic acid in avocaod's, olives and olive oils. These all point towards plant sources. And no wonder a Mediterranian diet might be so good since all these foods are also very present in that way of eating. Catechin with the gallate moiety is a sirt6 inhibitor, so I wonder if EGCG wouldn't counteract fucoidan and be a bad thing for longevity. I believe that green tea is good more for health span than life span. Epicatechin I believe from a study I read is less of a problem than catechins with the gallate. Alan Everett Alan Everett 1 year ago One of the big questions I've had about hyaluronic acid (and a number of other supplements) is the proper dosage. Your charts clearly show older people have much less hyaluronic acid than younger people. This reduction is true for a number of other molecules in our bodies as we age. You state that older people only have a quarter of the hyaluronic acid when you're above age 60 as you had when you were much younger. The charts also indicate as you go above 60 the levels go even lower. Since I'm 72 it would seem that I would need a significantly higher dosage of hyaluronic acid than a person who is in their 30s. Ideally we would have some sort of test so that we could use to establish how much more to take but I still am going to guesstimate and take more than you are taking. We are in new territory here but in other areas studies have established normal ranges for various molecules in our body I am making this judgment to increase my dosage based on the idea that there does not appear to be a lethal dosage (LD) that anyone has found using exceedingly high doses of hyaluronic acid or collagen. This lack of a LD is not true for some of the other supplements so caution is advised when trying to go up to higher dosage for other supplements for older people. We (my wife and I) are beginning to try and objectively measure what is happening to our skin. We just began taking pictures every Sunday and storing them so that we can look and see if improvements have happened over time. I am excited to see what other molecules you are going to talk about that will be helping the skin. You previously have discussed collagen well over a year ago but back to our story. About 5 months ago, my wife (age 74) and I (age 72), begin taking both supplemental hyaluronic acid and collagen (Doctor's Best Hylauronic acid supplemented by various types of additional collagen supplements we got off the sale rack at our Pharmacy or big-box store). It took months before we noticed an improvement as we are using the dosages that you have mentioned, that we now believe are to low for older people. We believe that these lower dosages are set for younger people. ie we believe we were underdosing ourselves. There is no question at all in my wife's instance that her skin has drastically improved. We intend to double our dosage next time we order refills for our hyaluronic acid supplements to see if we will get even greater Improvement based on the rationale I've set up set out above. Prior to beginning this hyaluronic acid and collagen supplementation my wife's hands and feet were so dry and cracked that sometimes the cracks went deep enough into her skin that they began bleeding. She has had numerous consultations with dermatologists and there was some improvement but the condition has persisted for over 20 years. The slight Improvement from the dermatologist was nothing like what has happened over the last month-and-a-half. All the dry cracked skin is gone and her hands feel like they did back when she was in her twenties. This all occurred while she was working at her nursing job and having to wash her hands multiple times a day. She had given up any hope that her skin would improve. She is now excited about our supplement stack. We also believe we're noticing improvements in the back of our hands which have thinned out skin on them. What lead us to begin photographing our skin is that we believe some of the age spots are getting fainter and going away and hydration is returning to these thinned areas of our skin. The jury is still out on the age spots and thickening of the skin but hopefully the photographs and chart notations and increased dosages will tell the tale in the next few months. We are also taking other supplements like nicotinamide riboside (NR) so we can't say 100% sure that it's just the hyaluronic acid and collagen supplementation doing this for our skin. Whatever it is, our skin appears to be getting younger or possibly we can just say it is improving greatly. (Also both of our hair is returning to its normal color which we believe is related to the NR supplementation. While hair grows out of the skin we believe the hair color change due to our NR supplementation because other people are reporting hair color changes. These hair color changes have been reported by people who are solely taking NR or NMN.) Hylauronic acid and collegen are also involved in cartilage formation in joints like our knees. After about five and a half months of taking hyaluronic acid and collagen we believe we've noticed an improvement in our knees but we haven't figured out an easy way for Lay people to measure it or to chart it to determine this for sure. In mouse experiments involving other supplemental drugs, they have proved improvements in knee joints by taking X-rays and actually measuring the thickness of the cartilage. We have approached my orthopedic surgeon to try something like this but he thinks we're nuts. He is been replacing people's knees for well over 30 years and didn't want to hear anything we had to say though he was very kind and polite and took some of the studies that we brought with us. We are very convinced that we should take higher dosages than younger people. As stated previously we have planned to up the dosage. Your previous presentations over a year ago are what convinced us to give this a try. Thank you so much and keep up the good work. I hope others will join us in trying to photograph and chart observations about our skin and joints so we can objectively prove these changes. I would love to hear what you think about our dosage hypothesis though I realize you can't be too specific as it would be considered a recommendation. Maybe some laboratory person somewhere will read this and begin the tedious process of measuring hyaluronic acid levels in younger people so we could establish a target range for our supplementation. I take HA but I am using Jarrow and I'm not sure that it's high molecular weight HA. I also take marine collagen and eat a healthy diet with lots of vegetables. I think because my body absorbed water and my inner ear structures became swollen. A year later, I tried 100 mg every two days, so effectively half of the dose. The same thing happened. I had an experience where I broke-out with crater-type bumps - all over my back, at 10 years old. It was extremely devastating. The dermatologist prescribed me Tetracycline, but it hardly phased them. Well, I loved being outside - So, I decided to start sun-bathing. To my surprise, the acne started to clear-up. I just started back on hyaluronic acid yesterday. I want to see if I will get the acne, again. If I do, I'll just take vitamin k2/D3 supplements, and get out in the sun between 3:00 - 4:00 pm, as this is the best time to not be as exposed to UV rays that are damaging. I need to use gloves when working in the yard and with heavy tools more because my skin is thinner and more prone to cracking and abrasions than when I was younger. At 200mg per day, it took 2 weeks before I could notice a difference. Trouble is, it causes the skin to retain more water than is necessary. I ended up getting rashes and had to stop. Some of the rash became infected with staph, and left scars on my legs. I'd do it again, but at 100mg on a 5 day on, 2 day off cycle. No real skepticism or meaningful critique here. Rather disappointing. I laughed out loud when said you would be taking HA. At 29 yo, that’s just flat out ridiculous. Someone over 60 maybe, perhaps, though I’m betting what is sufficient dosage is likely not well known. No justification for spending thousands in your 30’s and 40’s on this. Im 61.5 I have just started taking the collagen in the am@ 8g with my lemon water. I wanted to add in the HA but get nervous about overdoing so this weekend I started with 50mg on empty stomach with the collagen. then waited 30-40 min before my next round of supplements. was ok . Monday took 50mg of the HA ( double woods brand) and then 20 min took my L Tyrosine and R ALA which also have to taken on empty stomach - wow had a reaction. Heart racing feeling really weird. I've basically gone "all-in" and take NMN, Resveratrol, metformin and another 6 or 7 supplements, plus exercise nearly everyday, do intermittent fasting, meditation, etc., etc. My guess is I live to 251. 😂 Actually, I realize we're all Guinee pigs at this juncture with precious few conclusive long term studies performed on humans using some of these newer molecules/supplements. But for those over 50, I'm thinking, "Why wait?" After a long life, it seems like Vera is saying we can live to an older age if we are taking the whole list of DoNotAge products ?! or is it that we are more likely get a super bug or fall under a bus. Most people are living apparently without anything that suppresses the damages apparently natural to us, our lives will become senile. So after extending life and health what is happening, why is the battle eventually lost. I myself at 75 struggle with various inflammations arthritis but when we are visibly old we are written off by younger people who treat us differently when we look old, we are thought not to be able to contribute, we may not be able to be lively or properly useful even though as I can see many people are handicapping themselves with obesity and by seeing their life as being over after they have had children maybe they are bored long before they should and stop wishing to grow! Even now the products people like me are taking and our wish to be strong healthy and creative for as long as possible we know we can extend health how we look and our creative enthusiasm and feel valid. The hospitals are still full and doctors look askance at any suggestion that we can be more more healthy by taking NMN which has so obviously made it possible for me to renovate my letting flat being up ladders and doing most of the things my age group can’t, and even when programs come on the BBC lately they only mentioned metformin and even that with some hesitation. The Hollywood stars are the only ones believing in it “ out of desperation” for further work. Any way this is my basic feeling that belief in substances such as we take most are cynical about. I have so far not been able to convince anyone at all despite the fact that I look ten years younger than I am and that no one I know of my age will get up a high ladder and repair a ceiling and paint it, I’m female by the way.
