If you’ve been following Dr David Sinclair, you know he updates his supplement protocol from time to time.
And in each interview, he gives one piece of explanation to what he does.
And certain supplements that appear in one interview, he does not mention them in other interviews.
Sometimes it’s hard to follow his protocol today.
Since, I know Sinclair’s work since 2007, before he went public in 2019.
Here, I have collected for you from a variety of sources, including Sinclair’s book, trying to figure out EVERYTHING he takes today and how much from each supplement.
And, you’ll find a very spicy comment from Sinclair about resveratrol.

Great video! Thank you!!!
After having watched all your videos I'm still insecure about how much resveratrol to take (30-50mg or 500mg or even 1g like Sinclair does??)....Hopefully your forthcoming videos will help finding the right dosage...
As far as i know wheatgerms contain much more spermidine than the sources you mentioned.

I just ran across this channel today (Feb. 6, 2023) and it seems pretty good. I'm a fan of Dr. David Sinclair and Dr. Brad Stanfield, and I've attended two high-quality international conferences on longevity. Almost all this stuff is quackery and snake oil, but perhaps Rimon's content is not.

This is another very informative video. I just recently found your channel and really enjoy the content, especially when you dive into the details of Sinclair's comments and the studies.
Since this one is about supplements, have you considered doing some videos on liposomal supplements? A lot of these longevity substances have very poor bioavailability. Using a liposomal delivery system dramatically increases the amount that gets into the bloodstream (almost all of it from what I've heard) and also facilitates entry into the cells where it is needed. I'm in the process of switching to liposomal (or phytosomes, which is very similar and does pretty much the same thing as liposomes) supplements where they are available for the stuff I take.
One last thing, Metformin is mentioned in this video, and that is not typically available to people. You've also mentioned Rapamycin on your channel I believe as a longevity molecule. It turns out there are natural mimetics of those substances. On that reasoning I added ashwagandha, EGCG, and ginseng to my stack. See the study below for scientific support:

Just started resveratrol (for 1st time) at night after a meal, with coffee with the 2 tsp of olive oil and coconut milk. Not as bad as thought it might be. (after watching one of Sinclair's videos, ad lib.) Have to wait and see literally, results. Read about coffee as antioxidant. Also have read that it stops vitamin absorption. (Hopefully just slows time of absorption.)

How about this study concerning A-HgM (Metformin f.ex.) exposure in patients with T2D was associated with reduced risk of MS in patients younger than 45 whereas in patients older than 45, exposure to A-HgM was associated with an increased risk of newly diagnosed MS, particularly in women. Would this then also apply to Berberine?

When talking about vitamin K2 supplement dosage it’s important to be more specific on the type. I believe he was talking about MK-7 K2. You also have MK-4 which in most supplements is dosed higher than MK-7.

I would really like to clarify about Quercetin - it’s not safe to take it every day, it can damage healthy celks, and I dont think Sinclair is doing it everyday either….the study that he brings up (and I can look it up) used quercetin in hit-and-run fashion meaning 3 days in a row then 6 weeks break. Because there is no point to keep taking it everyday - once u remove senescent cells.

I'm a little surprised David takes so much of so many of these compounds. Most of them are synergistic to the point of actually amplifying the effects of one another.
The effects of Metformin and Aspirin, taken together, is more than the sum of each taken alone.
The effects of Metformin and Resveratrol, taken together, is more than the sum of each taken alone.
Metformin, Aspirin, Resveratrol and Quercetin are all synergistic, with many overlapping effects.
I have taken all of these, and in similar doses (but 1 g Metformin per day) but only for 4 weeks, as a purge. I wouldn't want to risk doing this long-term.
The only one I take every day is Metformin and that's because it's the only one of these compounds that has been proven to extend human life and has an excellent safety profile. On the other hand, one study on rats showed that Quercetin increased the occurrence of tumours.
Starting Monday, I'll be taking the polyphenol, Pterostilbene together with NR as they seem to have a powerful anti-cancer effect when taken together. Pterostibene has 4X the bioavailability of Resveratrol so I'll only be taking 100 mg per day. I'll also be starting with sulforaphane and eating much more broccoli together with mustard seeds (trying looking that one up).
When I was taking Resveratrol and Quercetin (my purge) I mixed them together with soya lecithin, on a slice of wholemeal bread. Both the lecithin and the wholemeal bread boost absorption, especially the lecithin, which has a strong affinty with water. I also mixed in 500 mg of VitC which I had in paste form, in capsules. The Vit C and lecithin help to protect the Quercetin, the lecithin in the stomach and the Vit C in the bloodstream, so it has more time in your system before being metabolised.
Anyway, thanks for keeping us up to date.... really appreciate your videos!

Great video! Just one thing: Taking drugs like Metformin is not natural and not healthy, it has side effects. If you do not eat sugar, you do not need Metformin! I do low carb - high fat - medium protein and IF and most days OMAD. Do sports in a fasted state. Take Pterostilbene on my resting days, not when doing sports.

Hi Rimon,
Do you address, or can you address, Berberine as an alternative to Metformin?
My doctor is not really up on longevity research, and she refused to prescribe Metformin to me. However, Berberine is touted as a natural alternative to Metformin, with many of the same benefits, plus others. Of course, I purchase Berberine online, so it's more accessible.
Thanks in advance,
Chris

Why does Dr Sinclair still take such large doses of Resveratrol even though in his own studies low doses worked better for longevity? In one of his recent interviews he mentioned that he now takes Resveratrol every other day.

So am I to understand just to be clear that he takes virtually all of these supplements at the same time or around the same time in the morning? I don't hear him or yourself mentioning that he takes anything later on in the afternoon or evening. (I heard him mention some people take NMN at night time). Just curious, is he taking D, K2, Resveratrol, Quercetin, Fisetin, NMN, Spermedine, and Tri methyl glycine all in the morning?
Thank you!

I saw a video by Dr. Berg where he stated that you can take 10,000 IU's of vitamin D3 with 100mg of vitamin K2 (MK-7 form only due to it's increased bioavailability) and not be concerned with calcium build up in the arteries, therefor you might be safe doing that.

Hmmm. When listening to this guy, he comes across as being envious and condescending when Dr Sinclair may adjust the use of what he may be taking.
It sounds like jealousy.
Isn't testing and adjusting the supplements part of assessing what the right level of dosing and compatability of products to produce the best products? He is experimenting on himself, so why is that a problem?
I don't get the reason for the snide comments with the tone of, "....well, he hasn't perfected it yet, has he?". It's work in progress.

What is your full name? What are your credentials? Are you a doctor? Do you have a Ph.D? A master's in medical science? Do you have any credentials? You make it look like you have interviewed David Sinclair and others, but I know -- from having followed Sinclair's work (who has a Ph.D., worked at MIT, heads the anti-aging program at Harvard) that you have lifted video form from other interviewers' work. You make statements about studies, but do not show their sources. In other words, nothing that you say can be validated. You may not be a fraud, but you give anyone who truly follows this work no reason to believe you. And the incredible arrogance and narcissism of calling yourself a Messiah!

He also takes a statin for a genetic predisposition for high cholesterol. This is strange because he eats coconut yogurt, which is high in coconut oil, which is well known for raising cholesterol.
Also, the 2008 Cell study mentioned only indicates improvements in longevity in the EOD feeding and HC diet cases. In fact, in the SD (standard diet) case, lifespan was decreased with low dose of Resveratrol. (Fig 4, graph E: https://www.cell.com/cell-metabolism/pdf/S1550-4131(08)00182-4.pdf)

This is a fantastic study and thanks for your breakdown analysis. I totally agree - and this now explains past studies that indicated that only 5–10% of all cancer cases can be attributed to genetic defects, whereas the remaining 90–95% have their roots in environment and lifestyle eg smoking, diet, alcohol, UV, pollutants, infections, stress, obesity, physical inactivity. Looking fwd to more on human studies too and your analysis. I will share this with my friend who is a genomics principal scientist.

These subjects are soo hard to explain, even to stem field students. I admire how you wanted to share this information in a more "people in general friendly" manner. Amazing how many aspects of aging is in our hands. I wonder how technology, if any, have a impact in our epigenome, the constant use of our phones and social media I think can make a impact in some way. what a great video

Great video, thanks. So the primary takeaway is that David Sinclair has demonstrated that epigenetic damage is more likely to be causing gradual aging than gene damage.
The secondary takeaway is that Yamanaka factors can potentially reverse some of this epigenetic damage, but are too dangerous to use, presumably because if you overshoot, you end up converting everything to stem cells..?..
So, a few questions:
1. How were the Yamanaka factors controlled in the experiment, so as to unwind the epigenetic damage without damaging the mice?
2. How will this affect the direction of research? Will it focus on how to safely use Yamanaka factors, perhaps by only using some of them? Or are there other methods that show promise?

How big of a roll does epigenetic tagging play with cancer versus genetic mutation. I worked as a health physicist and our story was the cancer being caused by generic mutations. But the fact that many people get the same types of cancers means it could not be due to strictly random mutations. Epigenetic differences could be a better explanation for how cancer types are so reproducable.

Great presentation as always nicolas. It makes sense now that the "epi-" in front of genetics meaning on top - may actually refer to these "tags" you refer to.
I am always ambivalent on topics involving NAD precursors supplements. While my biochemistry is admittedly pedestrian, there appears to be perplexed findings on their use on present literatures (particularly B3/NMN/NR and or even Metformin). Perhaps food for thoughts in the future?
many thanks. live-it-forward, AW.

Also, I know nothing about the NMN situation and Dr. Sinclair. This is merely an evaluation of his largest scientific work.

Would love to hear more on point 2. I mean it seems obvious to me the basic nut has been cracked and it also seems silly a 98 year old can't just roll the dice if he chooses to do so, but that's what regulatory capture will get for you. Gotta love corruption!

The Epigenetic cause of aging has been shown in multiple ways. Working with fish, this evidence has become more specific. Even perfect RNA cannot make protein/peptides/hormones without having all necessary amino acids. The current evidence points to a lack of amino acids due to changes in the microbiome. It looks like their may be a universal overgrowth of parasitic bacteria over time. Humans can have trillions more of specific gut bacteria as they grow older. After all, old people appear to be starving...

Would you consider a video on a common age associated disease such as osteoarthritis?
I'd be very interested to know effect diets (such as anti-inflammatory, sugar-free)
Supplements (collagen, UC-ii, boswellia, glucosamine, vit d, turmeric, etc)
Red light therapy
There's a lot of information online. But I wonder how much is back by science.
I'm desperate to help my mother who is struggling with hip pain from osteoarthritis

Your exposition of Sinclairs ideas and approach is very clear.
I like to see more from you with this quality.
The thing I wondered about is how equal the epigenetic changes of aging and the experiment were.
And how equal the result of "repairing" those was to the none-aged ones was.
What's unclear to me is if the epigenetic clock is a systematic change. Or if it's just a gross chaotic change in one direction.
So, a video about that would be appreciated, if this is someting the researchers even know. I suspect they don't really know, or they would have been more clear about it.
I noticed that you said nothing about that the mice being treated with anti-aging looked younger. And if they lived longer than damaged mice which didn't receive treatment. (Of course there would be some of those?)
Until I see some mice actually getting younger, I don't really belive that Sinclair has proved his thesis. Making mice older isn't really the problem!

Epigenetics is still a new word, a new idea for a lot of us, I just heard of the word about a year ago ,it fits in with many of our beliefs ,whether science or religion, a healthy lifestyle and healthy beliefs and memory leads to a healthy future, the memory part is in your young stem cells, that your parents forgot to freeze ( cryogenics) when you were young born, if you want to stay young, think young, how do you think placebos work, a positive mind is a young mind, Epigenetics, your genes are carryovers from your grandparents ,ancestral and environmental, The environmental part and that of the mind included, brings us closer to moments of notice to change in epigenetics, What is the epigenetics of a virus in terms of time compared to us humans, viruses are us, evolution, nature and memory, and then there’s biology ,virology and gene editing, we can actually tweak epigenetics in our favor, but the virus is faster, mutation, that’s epigenetics in a heart beat, you can’t beat nature, you can work with it, it’s in the genes, memory, fractal memory, does the spike carry the memory of the whole virus, mRNA to the host cell? Your playing epigenetics when you take the vaccine, pay attention to the telomeres, epigenetics

Thanks for the excellent breakdown. It was very interesting, indeed. I wonder what will happen when ageing becomes reversible. Imagine what would happen to the world's population. Or will the magic only be available for the happy few? Food for thought surely! Keep up the good work!

For me, the implication is that the evil powerful people of this world will be here far longer than we can bear. Another thing I noticed - what about the anti-inflammatory drugs role in aging? You didn't list them in your final three points, but they logically slow down the natural, yet unnecessary DNA damage.

That jest aside, do we know why the mice went blind? And if so, was there retinal regeneration. Or were there more simple corneal/lens issues? Either way, not a bid vid overall, well done.

Anyone knows how these Y T F (Yamanaka transcription factors) work? I mean what do they do, mechanistically? If they remove wrong epigenetic tags and (I assume) add original/healthy/correct ones to a) curb incorrect gene expression b) re-iniciate a robust/right/base gene expression - then- how do "they" (these 3 proteins) know how to do it? I mean how do "they" know which tags are supposed to be corrected? "they" must reach for some kind of a base "template" where that information is being stored...

Like all scientific research this should be taken with a pinch of salt until the results have been reproduced by other independent researchers.

Aging seems to be a series of mistakes happening within the body, overload of information, energy and cleansing required to sort and heal. Fasting, reduction of toxicity, reduction of stress, while obtaining energy from sincere sources, for instance the source of energy life maintains being able to produce, do not kill the plant, animal, or being, thus a possible reason the big ass goat/sheep evolved to that point, to show people its possible to obtain life force from the milk, the fat, the moss, the fruit, the herbs, seeds, light, air, water, vibration while allowing life to be. Roses have a beneficial conductive ability however that may be obtained while allowing the rose to live, thus the possible reason of roses with thorns and without thorns, its the electrical capability, the rose petals, the humidity above the rose, the scent, the air. Will man evolve to become a complete breatharian and or capable of harnessing electricity to support life without harming or interfering with life? Are the 120+ minerals we need primarily to charge and align our positive and negative charges to balance each cell and support life force to activate and maintain our innerchi?

i'd say the biggest problem in science of ageing is that people seem to be looking for that one single key cause or holy "knob" to turn it off or even reverse it and seem to be overlooking the possibility of it being managed by a larger file or system of many factors counted in at once.. a system that is only as strong as its weakest part.. something that is very sensitive to all possible aspects of lifestyle as a whole.. maybe everyone who ever thought he found that knob was right but he didn't feel like it because they've all expected their knob to be the master one and they were purely on a quest to find that one, not allowing themselves to be satisfied with anything less while nothing more superior than what they found might not even exist, only more and more of those they already found a lot of..

I’ve been taking nmn 450mg for a month and I noticed a spike in my energy straight away, work outs are much more enjoyable, deep sleep and dreams, skin is noticeably softer (I am 54)… went to a function with friends and family recently and two people asked what I was doing as I look younger! So I am hooked, I believe incorporating daily exercise and good nutrition is part of this as they help to generate the effects even more. The Pulse is a good Chanel to follow on this subject.

Please can you discuss the role of NMN in rescuing female fertility on the next one? I know there's not much research into this area as yet, but despite the impact on reversing ageing on mouse fertility, it's being ignored atm. If men had menopause I'm sure it would be a different story! I'm taking NMN in the hope it reverses menopause temporarily and boosts my egg health. I'm also taking tmg and several other fertility boosters. I'm on a low carb diet too to reverse insulin resistance.
Btw are you aware of the FDA ban on NMN as a supplement in the USA? Apparently it's being withdrawn from USA shelves on March 13th and recategorised as a drug. Not sure what that means for NMN takers in the UK as yet despite reassurances from various companies. I'm following a GP who's wife had her menopause reversed on NMN. The current research shows the ideal dose of NMN is 600mg daily and powder is usually cheaper than capsules. You can Google the info on dosage/trials etc. The info is on YouTube too. I also follow a channel called My NMN Experiment which posts lots of NMN studies that you might find useful. 250mg seems a very low dose to me but as you know it is an expensive supplement. You might want to add Apigenin or dried parsley or celery seed to your protocols to lower your CD38 levels to get the most from your NMN (CD38 increases with age and it lowers NAD). I hope it helps your parents. My dad has prostate cancer and I had a thyroid issue myself that I healed myself whilst reducing thyroxine, against medical advice....gradually! Not suggesting anyone should follow my example, but it can be done if you heal your body.

Thanks for the info! I've recently come across Pycnogenol and am starting to take it. I've read some amazing benefits, like it increases hyaluronic acid levels in the skin up to 40%! It also helps with blood circulation. Keeping the blood vessels on the face healthy and living is important to aging gracefully.

Very useful information especially when there are so many doctors promoting their supplements & diets! Hard to know which is best & right for one or what may be just hype. Thank you for investigative reporting! PS. Maybe your father may want to try the red light therapy on his feet for his arthritis to see if it may help as well?

I’ve been taking NMN for about three weeks and the first week was terrible. No energy, tired, sluggish- week three it’s finally got better! I feel rested in the morning, got energy during the day and im not wiped out after the day at night. It’s not the big burst I keep hearing about more a steady and consistent energy.

I take 500mg of NMN (based on human trials results) on Wednesday and Saturday mornings hours before eating my first meal at noon. Also I take resveratrol after dinner on Wednesday and Sundays, especially if there’s oils or fats in the meal. I was 56 when I started 4 years ago, feeling great so far, hair seems to have stopped greying 🤞

I agree that current research, in nutrition, shows that low glycemic index diets are beneficial, but I wouldn't, personally, try the carnivore lifestyle. There are so many antioxidants and fibers in whole grains, fruits and vegetables that both reduce cancer risk and support gut microbiome diversity. Nevertheless, this was a fascinating discussion and I just love the exploration of different ideas in emerging research on your channel! Very interesting topic!

Thank you Clare for your investigative interviews.
My understanding is to take resveratrol with NMN, this is the advice I have heard from scientists studying the area of epigenetics without direct monetary gain from marketing their own supplements.
Do you have any info or thoughts on this?

People said take NMN first thing in the morning, empty stomach. But I have to take my thyroid pills, first thing in the morning too. So can you suggest, which one I should take first? Thanks! (I'm in the process of ordering what you suggested. Hope it really work!) 🎉

If your mom has a Thyroid problem, I'm seeing it's hypothyroid or diminished Thyroid activity in response to normal or high levels of TSH production, I would suggest she take selenium daily. Not overdosing, but just taking the standard dosage but making it a daily habit. Copper and zinc are also helpful in helping to maximize Thyroid hormone activity. This is something NMN is not so effective for.

i wonder if there is also an element of a placebo effect on yourself and your parents. drug studies often report something like 30% improvement of symptoms in placebo groups (versus maybe like 70-80% improvement of whatever the actual drug is)

Clare, with all this talk about blood glucose levels, have you considered wearing a continuous glucose monitor for a few weeks to actually track it? There are several on the market. Levels comes to mind. Zoe offers one as well if you agree to be in their study/ies.
I just think, rather than say you “feel” like your blood glucose is this or that, it would be more convincing if you know.

I really wish this channel would feature guests who don't have a commercial motive. I take NMN/NR, but I get my information from credentialed sources (reputable MDs and PhDs) who aren't selling the supplements. Same with cosmetic products and devices. It's just more credible to rely on evidence-based research and get information from people who aren't promoting their products.

I like supplements, but EVERYONE should read "Fast and grow young". Our bodies are incredible miraculous creations with the ability to heal itself! All of todays "Dis-eases" are created by our diet. Our food lacks the nutrition it once had, and even 'organic food' has toxins from the soil. Fasting has long been doen throughout many cultures, and even animals instinctively 'fast' when they're sick. The book is fabulous and very eye opening.

Please run an nad test as soon as possible. Ideally you would run the test before the supplements, but you should do it soon to keep track of the progress. At your age your nad level should be at half of a teenager, so if you take the test now you can compare against that. I would be very interested i hearing your results. Also, you want to consider taking a biological age test, your biological age should decrease as you take NMN and TMG. If you take that test also please share your results.

Frankly, I'm on INFO Overload. I have Spinal Arthritis which generates daily Chronic pains and Terrible stiffness affecting my entire Body and Psyche. I take far too many supplements ....the list is long but does include Berberine, Turmeric, Ginger, Cinnamon, Bromelian, Zinc, Ds, Cs, Es, Mgs, K2s (with D3s). Wish I could say there is improvement. No Can do. Just had 3 recent MRIs done (Cervical, Thoracic, and Lumbar), the arthritis, the Disc Degeneration, sites of stenosis continue to increase. These conditions have been experienced for decades having started in the Lumbar Spine /Hips/SI joints and Transgressed up the spine to the top of my head. So, I seek "MAGIC" (there is no Surgery to help me per 6 surgical Consults). Like BIG PHARMA, I am increasingly leary to puruse even more Supplements (They are expensive too). I'd love to see what this doctor recommends. THX for your time to make this video.

If you can exercise u do not need to take berberine..in fact it attenuates the stress it puts on u and lessens your vo2 max...plus it was just found out that when NMN goes through the liver porthole it is changed to nicotinic acid..you are better using nicotinic acid that is a fraction of the cost and it does support the nad salvage pathway

It's a very interesting talk. I'm wondering, she mentioned the carnivore diet and I just have seen opposing data to the health effects of that diet. First of all, you have the production of AGEs, you have the production of TMAO by gut bacteria in meat eaters that leads to cancer and heart disease. Next, you have data showing the efficacy of the vegan diet in reversing aging. Bryan Johnson is a perfect example. He takes epigenetic tests to see changes in his biological age in response to everything he eats, and he says the prime diet is veggies, nuts and fruit. He has invested millions into optimizing his diet.He is vegan and he got that way because of the tests he has done on himself showing that meat is not a good option for someone looking to reverse aging. There is an insulin connection, there is a lot to do with blood sugar, but she's not differentiating simple vs complex carbs. Simple sugar is toxic to the liver, and should be avoided at all costs. Complex carbs are not going to cause the same blood sugar effects and there is more to it than just avoiding carbs. Meat is high in protein, which negatively affects the sirtuin pathways and leads to accelerated aging. Low protein according to Dr. Sinclair is the best option for reversing aging so that fact goes contrary to what has been said in this video. Berberine is also great but why are we avoiding mentioning the elephant in the room, ALA, Alpha lipoic acid, which has been shown to reverse cancer and not only stabilizing blood sugar but actually eliminating diabetes and reversing peripheral neuropathies in diabetic patients? And it's better than berberine, which is a weak form of metformin. ALA has been shown to work better than metformin. It would be nice to hear her thoughts on these subjects because the data shows the carnivore diet is not a good option and I'm very surprised to hear that. Hormonal optimization too is interesting to me because I for one have an hour glass figure and no visceral fat and I'm eating a vegan diet which is higher in complex carbs and fats and low in protein and so this idea that somehow carbs will cause this increase in visceral fat is untrue to me. It's true for simple carbs only.

In March 2022, I decided that I was sick of being 16st. I did these things
1. Cut out sugar
2. Cut out carbs
3. Began a carnivore OMAD LIFESTYLE!!!!!! This is not a diet it is a life choice.
I am now a healthy 10st. ( Though BMI said I should be 7.5st. Ermmmm no way I would look terrible🥹)
I take NMN and TMG as-well as other supplements. At 60 I feel better than I did at 30. I have energy confidence and a new zest for life. PS Love the channel ❤

The answer to the question about these supplements feeding cancer did not address the actual problem. Of course, doctors will not give these supplements to someone diagnosed with cancer. The issue is, taking these supplements BEFORE knowing of cancer diagnosis and unknowingly feeding cancer by taking these supplements. Would be ideal to have this addressed.

In 1982 Pearson & Shaw in "Life Extension, A practical scientific approach" suggested caution in abruptly stopping megadoses of water soluble vitamins because of the potential of causing symptoms of vitamin deficiency if cells and tissues acclimated to high levels of vitamins are suddenly deprived of them. For this reason I doubt that "cycling" a water soluble co-enzyme like NMN is a good idea, at least not without a gradual taper off the dosage over a period of two or three weeks. For the same reasons Dr. Seranova recommends ramping up dosage when starting a new supplement, when quitting I think it advisable to ramp down the dosage over time as well. Cells seek to maintain Homeostasis, and abrupt changes mess with that.
