In this episode, Dr. David Sinclair and co-host Matthew LaPlante discuss why we age. In doing so, they discuss organisms that have extreme longevity, the genes that control aging (mTOR, AMPK, Sirtuins), the role of sirtuin proteins as epigenetic regulators of aging, the process of "ex-differentiation" in which cells begin to lose their identity, and how all of this makes up the "Information Theory of Aging", and the difference between "biological age" and "chronological age" and how we can measure biological age through DNA methylation clocks.

I am 27 and in the last few years I began to notice, that some of my peers are aging much faster than me. This observation caused me to often contemplate the topic of aging, and when I listened to Dr. Sinclair on Huberman Lab, I subscribed to Lifespan right away. My wife and I are 65 yo and we want life after 65 to be our active second life not a death sentence.
We plan to share this information to whomever will listen and become disciples of longevity.

Dr. Sinclair is a hero. We owe him a debt of gratitude. I have been following him and his work since 2016 like every single week. I'm so glad that there is a podcast now!
The only thing that I'm worried might derail this fantastic and groundbreaking research is the political turmoil in the US. We need to make sure that this research reaches fruition no matter what.

I have listened many of Dr. Sinclair's talks and interviews but this podcast is really well orchestra to enable viewer to better understand the subject matter. We also say in Hebrew "Lechaym" which means "to life" every time we raise our wine and cheers.

I remember about 20 years ago when I still went to school and we didn't have all this great information that is now available online, I often said "aging is just a disease, we could be immortal" and everyone said I was stupid and laughed. I've read the book, seen other shows with Dr. Sinclair and each time I'm a bit worried that maybe I will hear the same stuff again. Nope. Each time there's something new. Covid made me reconsider my life priorities and made me decide to go back to school and study biochemistry and molecular biology. I've read some of your works about the expression of 3 of 4 yamanaka factors in mice and it made want to focus my studies on trying to prolong human life / quality of life. Thank you! I feel like our current lifespans are a bottleneck to our civilization's advancement. Imagine what could be if our greatest minds could live and thrive for 30-40 more years in their prime.

Can you do a podcast on development of children? The acceleration of aging during puberty seems to be somewhat polar opposite to the idea of fasting during adulthood. What ideas come to mind when we say “skip a meal” to a 25-45 year old vs to a 7 yr old, 13 yr old, etc? How do you think about optimizing health in children that are developing in height, performance, etc? Thank you for all you guys do!

I first heard of the book, Lifespan, on the James Altucher podcast at the start of the pandemic. I bought the book and I talk about the concept that aging should be treated as a disease. This is all very exciting stuff! I would like to hear you speak on the ethical and philosophical ramifications of this. While of course slowing and reversing aging — and certainly preventing disease — sounds great, the idea of everyone living forever honestly sounds pretty terrifying. Not to be a total cynic, but it seems like the turnover of life is an important part of the progress of our species in so many ways. If people suddenly start living indefinitely, no longer leaving jobs or positions of power but just continuing to grow and hoard wealth, how will that affect upward mobility? As those people maintain their power for decades (or centuries?) longer, how much more stagnant could social change and progress become? How will it affect the global ecosystem and climate? What about people in developing countries who don't have access to even the low-cost suggestions you make? Could the ability to deliberately age or de-age a person be used for evil (as a punishment, for instance)? Again, I'm not trying to be overly negative, these just seem like very important questions we should be asking and addressing if this is actually where our species is headed, and I'd be interested to hear what you have to say about it.

This is amazing to listen to and see the interaction between the two of you. I remember a few years ago I was taking a microbiology and anatomy and physiology class at the same time. I remember the microbiology class was going in depth of the cell and learning about DNA, genes, and each section of the strand with how they link together with the histones. For some reason I became interested specifically with this and how X differentiating and mutation of cells leads to cancer and the aging of the body. The anatomy and physiology class went into some of this as well because it was the second level of the class which was more in depth of the physiology relating to the anatomy.

I went to my hairdresser yesterday, we are in Perth Western Australia, and was telling him all about the first podcast. He's nodding away, I know, I know he says. He rushed out to get his phone and he is listening to Lifespan on Audible. Fantastic! I'm really looking forward to this series. Thank you Dr Sinclair and also to Professor Huberman for your knowledge and making it accessible to the general public. Oh and I love listening to an Australian accent (even though it is very light).

Incredible, loved all of it..got some amazing new information for example the HbA1c and how Metformin can help. My levels were at 5.6% when I had my first Insidetracker test done last October so will be interesting to see what they are at soon. Can't wait for the next episode!

The wait is over.. My guru is Live. No words can ever comfort me to express my gratitude for all that I have received from life especially from my Guru “Dr.Sinclair”. I have been fortunate to have followed you since the past 4 years and all that I can say is that I am a 37 years chronological getting oldman but who looks like a 27-28 years young & health. It’s because of following you and your lessons on longevity I have been able to successfully reverse my biological age to live a robust & active life till a 100. As Dr.Sinclair once said “The one who will live till 150 has already been born”, I will do my best to be among the set of people who do. In the end can’t wait for your new Book and listen to the remaining 7 episodes of this podcast series. I bow before you for all that you have taught me.🙏🏻

Thank you thank you David Sinclair and co host for speaking so clear for us that are not on the scientific world. I was a mother at 44 years so my interest in keeping alive with my best quality possible as long as i can is a huge wish so i can share with my child many things in his adulthood. Hope as what you say, devices and everything else will be more accesible for us in the future, but we can start with what´s in our hands and that i have been reading/listening over the last years and it´s what you talked about on your episode 2 which i loved too. I´m excited for the next episodes!!!!!

2 things should be adressed:
How to balance fasting with geting enough nutrients and what foods are the best.
What are the foods/extracts that are similar in effect to the substances you recommend. Like is grapeseed extract a good alternative for Resveratrol.
I think this would be useful for people in poorer countries.
Also, a PDF at the end of the podcast with the main points would be awesome!

Thank you so much for sharing this with us! I’d very much like to also hear your thoughts about ASD, Dr. Sinclair. 1 in 44 children in the U.S. is diagnosed with an autism spectrum disorder (ASD) and this epidemic is global. There is no known single cause for autism spectrum disorder, some say it’s genetics, some say it’s environmental. most children have gastrointestinal, mitochondrial, sensory issues, inability to detoxify and the list goes on. These kids are born with ASD. Please share your thoughts on this disorder, is this related to early aging and can we fix these so called “dirty genes”. As a mother of an autistic 3 years old child, I’m eager to learn more about this and help my child as much as I can.

David Sinclair has helped me be a healthier and younger person since I was in back in 2020. I’ve implemented intermittent fasting and mild NAD+ boosting precursors like NR and NMN and I’ve been consistently using mineral sunscreen as of a month ago. I read his book Lifespan and I’m starting to read the book again it’s such an interesting read and learning about his family, especially Sinclair’s grandmother, Vera’s positive influence and motivation towards his life. I’m 20 now by the way

Complicated topic, I need to spend more time on it but you guys did a wonderful job explaining it to someone with no background. I was pretty skeptical on this idea and a lot of doctors online are too. All they need to do is listen to your podcast and they will have the background to understand. The better grip the average Joe has on this the better we will have of explaining it to those that don’t want to listen or simply not that interested and are kind of caught up in lifestyles that are killing them rapidly. This springs so much hope for my child’s generation, we can try to extend the life and the quality of it for those that are here with this knowledge and let us live out the rest of our life with a higher quality, by my child’s generation or his children’s children we’re going to see some really serious changes with this knowledge. I wish I could peek into the future 200 years from now then again at 1000 and 2000. We may survive this planet yet, all we need to do is stop killing our own through things like the poverty plate lack of healthy and hygienic sleeping conditions materialism and capitalism. If a rich person can build a brand new house after 15 years and tear down the old one why are we sticking people in homes so old they have mold, toxic mold? Greed… The root of all evil. When someone believes they deserve more than others because they have been fed the idea we injure our own species and ensure our demise. You can’t exactly blame the planet for trying to purge us now can you, not if we can’t get our shit together.

Thank you very much for doing this! Exciting and illuminating! I haven't read the book yet but I am going to. I am indeed confused about how telomere lengthening and hormone replacement therapy fit or don't fit into this information theory of aging. Likewise, some supplements may be hurting in excess amounts and helping in proper amounts (vitamin C and vitamin E). Ultimately, one thought has been troubling me for years: with knowing so much about the human body and understanding so much about genetics scientists cannot give clear-cut recommendations on how to exercise, what to eat and how to organize your life. I have high expectations that this series with bring order to this mess.

Dr. Sinclair, based on a comment you made, what's the reason people lived so long before the Biblical flood and started getting shorter lifespans after that? As a side note, THANK YOU!! I've been following your research for several months now and am beyond excited... It's like I'm see another life in front of me literally, and with youth. It's mind-blowing to say the least.

I think Matt asks really good questions but the interruptions can get a bit jarring at times. Can you maybe tweak your format a bit so Dr. Sinclair can finish explaining then just do a "going back to what you said earlier" just like Prof. Huberman does in his podcasts?
