I just recently found your channel and really enjoy the content, especially when you dive into the details of Sinclair's comments and the studies. Since this one is about supplements, have you considered doing some videos on liposomal supplements? A lot of these longevity substances have very poor bioavailability. Using a liposomal delivery system dramatically increases the amount that gets into the bloodstream (almost all of it from what I've heard) and also facilitates entry into the cells where it is needed. I'm in the process of switching to liposomal (or phytosomes, which is very similar and does pretty much the same thing as liposomes) supplements where they are available for the stuff I take. One last thing, Metformin is mentioned in this video, and that is not typically available to people. You've also mentioned Rapamycin on your channel I believe as a longevity molecule. It turns out there are natural mimetics of those substances. On that reasoning I added ashwagandha, EGCG, and ginseng to my stack. He started to take higher doses of K2-7 and asked for tests after taking this high dose of K2-7 and his valve showed significant decrease in calcification. I take 1gm NMN daily sub lingually. I have ordered 100 gm resveratrol and Fisetin on line and am in a quandary about dosage. I take Vitamin D 10,000 and Vitamin K2 90 micrograms daily. At 81 I want to stay on track. I read Lifespan and Super Human and have used NMN since 2018. I purchased a NMN Resveratrol mix on line for my son which he takes sublingually. This was because one supplement was easier for him to remember. My GP prescribed Metformin 1gm when I asked to keep my Blood sugar under control and it coincidentally reduces my blood pressure. Give my son 250gm Metformin. Advised him to ask his GP for a script. He has high BP and my father had diabetes so reasonable request. He My GP said he has been taking Metformin for years as preventative health measures. His father too had diabetes. Most of them are synergistic to the point of actually amplifying the effects of one another. The effects of Metformin and Aspirin, taken together, is more than the sum of each taken alone. The effects of Metformin and Resveratrol, taken together, is more than the sum of each taken alone. Metformin, Aspirin, Resveratrol and Quercetin are all synergistic, with many overlapping effects. I have taken all of these, and in similar doses (but 1 g Metformin per day) but only for 4 weeks, as a purge. I wouldn't want to risk doing this long-term. The only one I take every day is Metformin and that's because it's the only one of these compounds that has been proven to extend human life and has an excellent safety profile. On the other hand, one study on rats showed that Quercetin increased the occurrence of tumours. Starting Monday, I'll be taking the polyphenol, Pterostilbene together with NR as they seem to have a powerful anti-cancer effect when taken together. Pterostibene has 4X the bioavailability of Resveratrol so I'll only be taking 100 mg per day. I'll also be starting with sulforaphane and eating much more broccoli together with mustard seeds (trying looking that one up). When I was taking Resveratrol and Quercetin (my purge) I mixed them together with soya lecithin, on a slice of wholemeal bread. Both the lecithin and the wholemeal bread boost absorption, especially the lecithin, which has a strong affinty with water. I also mixed in 500 mg of VitC which I had in paste form, in capsules. The Vit C and lecithin help to protect the Quercetin, the lecithin in the stomach and the Vit C in the bloodstream, so it has more time in your system before being metabolised. (after watching one of Sinclair's videos, ad lib.) Have to wait and see literally, results. Read about coffee as antioxidant. Also have read that it stops vitamin absorption. I believe he was talking about MK-7 K2. I'm interested in the biochemistry of the metabolic process. This is what I found out about NMN. It sounds entirely plausible. BTW Apigenin is the extract essence of the chamomile flower, the same one used to make tea. I'll buy the R-alpha-lipoic acid too. I already take quercetin 500 mg and TMG. IMO resveritrol, spermidine, and Metformin have been discredited. Better to take no resveritrol than a high dose. I think the original research reached its conclusion based on an error. If spermidine works at all a handful of green peas a day should be sufficient. Metformin seems to have no effect on life extension. It has many drawbacks over berberine. I've had to cut back on berberine to 450 mg per day. that he mentioned in the 1:17 book let's hear it from him 1:21 what are you currently taking uh Doses 1:24 and such like 1:26 yeah well actually it hasn't changed 1:27 since I wrote uh lifespan 18 months ago 1:30 so page 304 1:32 see the list 1:33 now if you do consider to take these 1:35 supplements you wonder why does Dr David 1:37 Sinclair take these vitamin d and 1:40 vitamin K2 supplements in this segment 1:42 it explains why so you can make up your 1:44 mind about your health 1:46 I take vitamin D vitamin K2 K2 is a 1:50 really important one which keeps the 1:51 calcium out of your arteries and puts it 1:53 into your bones where it's needed so 1:54 even though Dr David Sinclair doesn't 1:56 mention vitamin D and K in every 1:58 interview it gives I guess because they 2:00 are more nutritional choice of 2:01 supplements than longevity supplements 2:03 on which is mostly asked about the 2:06 reasons he gave are solid and it's safe 2:08 to assume that it still takes them what 2:10 about the dose of those two supplements 2:12 I never found one interview where he 2:14 mentioned specific doses of those two 2:15 supplements a standard dose of vitamin D 2:17 is between 2000 to 5000 units and a 2:20 standard dose of vitamin K2 is somewhere 2:23 between 50 to 120 micrograms so I 2:26 estimate that his does fall within this 2:28 range the verdict he still takes vitamin 2:31 D in K2 and the dose is unknown let's 2:33 move to the next one this is his most 2:35 famous supplement Resveratrol 2:38 foreign 2:41 so let's go back in 2019 book he 2:44 mentioned I take one gram of any man 2:46 every morning along with one gram of 2:48 Resveratrol shaken into my homemade 2:51 yogurt does he still take the same dose 2:53 in 2022 in his very long interview in 2:55 his channel he confirmed I'm quoting 2:57 I've been taking one gram of Resveratrol 3:00 since 2004. so it's the same exact dose 3:03 the one he wrote in his book confirmed 3:05 of course if you follow Sinclair it's 3:07 not just how much respiratory takes but 3:09 also how now that has been evolved over 3:12 the years so I showed you in the book 3:13 that he mentioned in 2019 that he mixes 3:16 Resveratrol with yogurt about a year ago 3:19 we changed that protocol and then began 3:21 to mix Resveratrol with olive oil 3:23 however I was pretty surprised to see 3:25 that he also changed that protocol as 3:27 well let's hear from him what he is 3:28 doing now 3:31 um I've even tried I like to mix up my 3:33 diet like anybody else I've tried olive 3:35 oil plus a bit of vinegar and a Basil 3:37 Leaf in the morning it's like drinking 3:38 uh we call it salad dressing in the 3:41 morning uh that one lasted for about a 3:44 month I've given up on that I now like 3:46 this coconut yogurt that you've turned 3:48 me onto actually 3:50 the verdict one gram of Resveratrol 3:53 mixed in coconut yogurt in the morning 3:55 as a segue here if you have doubts about 3:58 adding Resveratrol into your supplement 4:00 routine don't go anywhere we're going to 4:02 attach this Resveratrol controversy 4:04 today the next two supplements are from 4:06 the same group the same family of 4:08 Resveratrol the polyphenols the 4:10 flamenols 4:11 [Applause] 4:12 foreign 4:17 explaining why he takes these 4:19 supplements 4:23 um so I've added 4:25 um 4:26 quercetin which is a molecule related to 4:29 Resveratrol which is also 4:32 suppresses the activity of senescent 4:35 cells the the one that I'm testing out 4:37 is physician f i s e t i n n because in 4:41 we showed in 2003 and 2005 in two nature 4:44 papers that it extends the lifespan of 4:46 animals small animals worms and flies 4:49 but nevertheless it's been now shown 4:51 that it's analytic it kills off the 4:53 senescent cells in the body at least in 4:55 mice probably in humans based on some 4:58 human data and so I'm looking at that 5:01 so this is the why but how much does it 5:03 take this segment is from a long video 5:05 in his channel 5:09 visadin and Kirsten half a gram in the 5:11 yogurt 5:12 the verdict 500 milligrams of quercetine 5:16 in phisidine every morning mixed into 5:18 yogurt probably now we can assume it's a 5:20 coconut yogurt 5:21 [Applause] 5:22 [Music] 5:26 the next supplement in our list is an MN 5:28 let's hear Dr David Sinclair explains 5:30 about this supplement and how much he 5:32 takes and so nmn is is what I take each 5:35 day I take a gram of it I know a gram is 5:39 likely to be raising 180 levels during 5:41 day 5:42 I also try to time it with my natural 5:44 circadian rhythm so NAD will go up 5:46 during the morning getting ready 5:48 um 5:50 but if I take it at night 5:53 what I find is that I'm actually 5:55 starting to interfere with my sleep 5:58 patterns in all the interviews he has 6:00 done since then it's still the same dose 6:02 one gram of any man in the morning and 6:05 it doesn't need to be mixed into 6:06 anything just with water so the verdict 6:08 one gram in the morning with water let's 6:11 move to the new supplement in Dr David 6:13 Sinclair protocol 6:14 [Applause] 6:16 spermidine 6:19 let's hear what he says about that 6:24 uh spermity there is you can buy it now 6:26 it's um there's a company that makes it 6:29 in pure form very low levels of gluten 6:31 and just the last few months I've added 6:32 that to my protocol and we'll have to 6:34 see how my numbers look on inside 6:35 tracker okay so that's not that's not 6:38 something you've adopted and you're like 6:39 I'm definitely sticking with it this is 6:41 I'm adopted and I'm testing it out to 6:43 see how it works I am and I actually I 6:46 advise that company it's the first 6:47 supplement company I am advising and I 6:49 did that because I wanted to look at the 6:51 human clinical trials and they they look 6:52 really promising as well how much of 6:54 that are you taking a gram as well okay 6:57 so from this interview it sounds like 6:58 this is the most recent experimental 7:00 supplement in the stack now what about 7:03 the dose this is important to listen to 7:05 this even though in this video Dr David 7:07 Sinclair mentioned it takes one gram of 7:09 pyrimidine later in the comment section 7:12 he corrected this dose he said 7:14 correction I said I take one gram of 7:16 spermidine but the active ingredient in 7:18 the capsules is one to two milligrams I 7:20 apologize for the confusion so the 7:22 verdict about spermidine is one to two 7:24 milligrams taking every day in the 7:26 morning this is what he said also in 7:28 this podcast now if you too consider to 7:30 take spermidine and you think okay this 7:32 is only an experiment he's doing on his 7:34 body maybe if there is a safer and 7:36 cheaper way to guess permittent and many 7:38 of my subscribers ask me how to guess 7:40 permitting from food this study measured 7:42 how much permadine in each type of food 7:44 and and ranks them from the highest 7:45 concentration to the lowest I'm quoting 7:47 from this study spermuding concentration 7:49 in Foods was as follows mushroom the 7:51 highest 7:52 88.6 milligrams per kilo of food green 7:56 peas 65 milligrams per kilo of food and 7:59 broccoli 32 milligrams per kilo of food 8:02 so as you can see there is a bunch of 8:04 spermidine in mushrooms but also in 8:06 broccoli and green peas these levels 8:09 coming from food more than satisfy or at 8:12 least similar to the levels of 8:13 spermidine that Dr Sinclair takes in a 8:15 supplement form and also there is 8:17 another certainty about the safety of 8:19 spermidine in those Foods 8:20 [Applause] 8:21 [Music] 8:25 now we are getting more into the 8:27 controversial stuff let's hear Dr David 8:29 Sinclair explain on why he used to take 8:31 TMG the reason that I take 8:33 um 8:34 glycine actually specifically 8:36 trimethylglycine is is actually to 8:39 counter what I think might be going on 8:41 with an NAD booster but the thing with 8:44 nicotinide mononucleotide nmn is that it 8:47 it has this nicotinamide group on it it 8:50 hangs off the the main part of the 8:52 chemical and it's the first bond to 8:53 break and so we see in animals and even 8:56 in humans that 8:57 the levels of nicotin might go up quite 8:59 rapidly after taking nmn or NR and two 9:03 two high levels of nicotinamide are not 9:06 good in part because 9:09 the nicotinamide gets excreted through 9:12 the kidneys and it's done so that 9:15 happens because it becomes methylated 9:17 into methyl nicotinamide but the concern 9:19 that's that's been talked about in 9:22 social media especially is is this drain 9:25 of methyl nicotinamide a problem the 9:27 methyl groups are are needed for the 9:29 body we need methyl for a whole range of 9:31 things including antioxidants 9:34 and so as a precaution I take 9:36 trimethylglycine so that I continue to 9:39 give my body a source of methyl groups 9:41 now I don't know if that's true people mention TMG 10:27 in his last interviews as he stopped 10:29 taking TMG your guess is as good as mine 10:32 but as we can see how he adds and Stack 10:35 Up supplements the likelihood that it 10:38 doesn't stop taking them unless there is 10:40 a very good reason or risk of taking 10:42 them and TMG is a pretty safe supplement 10:44 how much does it take he never said but 10:47 the standard dose hear him 11:10 explaining exactly that I try to take a 11:13 couple of Metformin pills for two 11:16 reasons one is that my family has a 11:18 history of diabetes and metforminism 11:20 very effective at treating diabetes and 11:22 even preventing it so I do that for 11:24 disease reasons but also because the 11:27 work of many Labs has pointed to 11:30 um not just animals but tens of 11:32 thousands of people in clinical trials 11:33 benefiting from that drug which seems to 11:37 enhance and mimic the benefits of 11:39 fasting now what about the dose so we 11:42 said in the book it was one Graham he 11:44 said it takes one gram of Metformin 11:46 every day apparently it changed a bit 11:48 with those in a recent podcast he said 11:51 I'm quoting I take 800 milligrams at 11:53 night but the edit that it does not take 11:55 me to form in the days before exercise 11:58 anyhow 800 milligrams in one gram is 12:01 very close dose so it hasn't changed 12:03 dramatically about the time in 12:04 government forming Dr David Sinclair 12:06 says that he takes it at night this 12:09 corroborates what I found with with my 12:11 own body in my own experiment with 12:12 metformin over the last three years I 12:14 found that metformin is very useful to 12:16 my body before bedtime because it 12:19 offsets the glucose stimulating hormones 12:21 at night melatonin and growth hormone 12:24 these two are natural hormones produced 12:27 by the body and they are increasing the 12:28 Sleep Quality they are good hormones for 12:31 anti-aging however one of the things 12:33 they are doing is increasing glucose at 12:35 night so personally I found what Dr 12:37 Sinclair I guess found himself that 12:39 metformin helps to reduce the blood 12:41 sugar at night but again it's a drug 12:43 it's not a food supplement and it's just 12:46 my experience and not a recommendation 12:47 to anybody the verdict about metformin 12:50 800 milligrams six days a week day off 12:53 the night before confusing if you're taking 13:48 Resveratrol on its face value there is 13:51 no reason not to go with Sinclair take 13:53 on Resveratrol but what is even more 13:55 confusing is that those studies
