All my life I took care on what to eat and exercise ...I’m Belgium descendent Born in Paraguay South America . Only ate when feeling hungry healthy vegetables green tea from Paraguay No wheat ... (turned out to be gluten intolerant ) So from 5 years old did not eat bread it made me feel bad , stomach gastric acidity.. so I didn’t eat it )... now I feel like 12 and I am 68 years old ... In our view, to really address aging academic medicine is essentially obsolete. The fact that the TAME study with Nir took ~4 years to get going says it all. So you know, we are older than you. In our view, your eye program has potentially very serious flaws wherein your actual thesis is correct but will fail after years of work. With this said we’d like to connect with you. Like we are obviously used to being impossible. I suppose at the time your test age was so much higher and you tried these drugs your hair was brown and the way it is today. Would they be able to live long enough to see humanity reaching another solar system? Maybe. Just because immortality might be centuries away, it doesn't mean we won't live long enough to reach it. 4 Reply Dan Dan 2 years ago (edited) I hope that in 20-30 years we will see the advent of cellular engineering, first of all gradually eliminating and reversing the aging factors. Eventually, we will be able to reverse and stabilize human biology at ~20 years old, perhaps completely so in 50 years from now. Although I feel current improvements are minor, I think that Dr. Sinclair is establishing some groundwork on that. Eventually we will have to revise social norms and regulations as this transitions to full genetic and epigenetic engineering at a cell-by-cell level in mature organisms. Perhaps in 100 years from now, it will start to mix with synthetic life components (which will be nothing like we understand by hardware nowadays). Therefore if there is regeneration of the first two (so called) cranial nerves then you are accomplishing a higher level outcome as opposed to the regular cranial or peripheral nerves which are well known to repair themselves spontaneously e.g. facial nerver palsies routinely recover. At my age 79.5, I am definitely impressed with changes I have experienced over the past 8 months of NMN use. I still continue with my others from The Life Extension Foundation which I have been using over the past 40+ years! He has multiple chronic conditions, chronic kidney failure (stage 1 now, was stage 3), chronic heart failure (various improved markers), and vascular dementia. This is an unqualifiable observed improvement in memory and cognition, and incontinence. At the risk of being blunt, basically he's not peeing and pooping his pants or his bed anymore (This is huge). However, dad is taking 2.4g per day (huge dose based on mouse studies). We started at a lower doses for 12 months @600 mg per day. We saw no meaningful improvement, and then went higher after a stroke. At this new higher dose dramatic improvements were observed after 16 weeks. He also takes 2g of TMG. Great review and analysis of the letter. It is sad that there’s doesn’t seem to be an easy way back for David Sinclair to return to being seen as defender of the people. His work so far has brought so much hope and now it’s overshadowed by this business decision that pits peoples desire for health against his company’s ability to make money whether it be for future research or simply pure profit. As a supplement, it still makes money for many but not the people and businesses that are used to making money through monopolization. I think there’s a big difference in making a product that people “can’t live without” (as a new convenience that can increase productivity) and patenting it for profit and making a product that constitutes a fundamental cure for a human disease that people will die without. I appreciate your perspective and thoughts on this. The emphasis on “FDA is good” appears to be the intended primary takeaway. The reasons are still unknown. There are definitely forces at play, and it’s certainly easy to start blaming David Sinclair as a greedy antagonist. Maybe that’s true. I don’t know. But also I don’t need to make a judgement on him. Part of this is the certainty that.the US supplement industry will communicate with the FDA on the issue (indeed, one other presenter right here on YT, Brad Stanfield, IIRC, states they have already begun the discussions) and may bring suit against the FDA, if necessary). Part of the industry's logic is the documentable fact that NMN has been sold long before Metro Biotech began their studies of NMN, or some derivative, as a patentable drug. If he had nothing to do with this and was against it considering he is part owner of the actual patent for this MIB stuff he had control over what happened to his patent lol. Me and my wife recently started NMN + Resveratrol at 500mg NMN + 1g Res upon waking. The first night we both had insomnia and for the following three days had pretty pronounced side effects. These included but were not limited too insomnia, dehydration, poor memory, bright urine, allergies, lethargy. My best guess is that this is caused by undermethylation and the immediate loss of all of our B-vitamins. I believe the fix could be adding TMG at some dose. I’ve read a lot of people having similar symptoms on forums. Yes, he is a co-founder of a company that could possibly benefit from the huge profits gained by their version of NMN, but #1 - that doesn't mean that he has a controlling interest, does it? Could be, we don't know. 2. When Resveratrol was attacked, he personally was attacked and it took years for him to regain his reputation. I can just imagine that this slowed reaction could be his caution from being burned before. 3. If you have been following his interviews and postings for the past year, you should note a more insistent cry for research funds to cover the huge expense of the work he does. Harvard gives him housing, not necessarily monies to pay for this research. This mostly, as I understand it, comes from private (very wealthy) individuals and corporations, and it appears with the financial instability of the entire world, these funds are drying up - rapidly. His lab needs money. Rather than judging the man and speculating what all this means, I, for one, will wait to see what happens. There is a two tier system where a certain substance can be both a supplement (usually much cheaper) and a drug (usually much more expensive). Case in point : NAC is both a very moderately priced supplement and a drug (more expensive) used in hospital emergency rooms to treat Tylenol overdose toxicity to the liver. Both co-exist. Question, of course, can be asked : why pay more for the same chemical ?? Another example is fish oil EPA and the drug Vascepa. Some would say these two products are Not the same. Vascepa has been chemically twerked to be slightly different, but the net result really is a higher concentration of EPA and that the same EPA amounts can be achieved at a cheaper cost with supplemental over the counter fish oil, so again, why pay more ?? Answer : to protect the interests of the pharmacistical industry, Not the general interests of the masses (the public). Look at it this way : it's an example of the tyranny of the MINORITY. His company is undergoing clinical trials for a different type of NMN in crystalline form to be sold as a drug. That is why he is saying that he has never been involved with the marketing of NMN as a supplement, as your company donotage sells. While I think it is generally safe it is significant to note that Peter Attia now recommends that his patients do not take NMN. Previously, he had not recommended it because no study has ever shown that NMN increases longevity, but he didn’t discourage it them from taking it. NMN failed the ITP trial and other labs have also show no positive effect. He based his conclusion on the new study that shows that there is possible risk that it could increase the growth rate of certain types of existing cancers. That has by no means been demonstrated, but given even a slight risk and no proven benefit, he changed his recommendation. My reasoning being is that David Sinclair has always been saying his goal for NMN is to be a fully regulated drug, preferably over the counter. So that means following the current laws on regulation, not sure why that would be surprising and also what would one expect him to say/do if that is what needs to be done. If the goal is to get the most people possible using it then that is simply not going to happen if it's only a supplement. The thing is the majority of the public and the Medical community will accept it's efficacy and thus use it if it's an over the counter drug and not simply a supplement. Maybe the majority of people's feelings about supplements will change in the future? I personally am the only one(aside from my wife), I know who takes NMN, my step Mom is interested in it but my Dad is disinterested and unconvinced of it's efficacy and my brother will try it but only if I send him some, LOL. People I know and met who are interested in longevity and overall health don't use and for the most part haven't even heard of it. I'm 68, I've lifted weights for over 40 years so I suppose I'm stronger and fitter than most guys my age. I'd like to say something about the energy provided by supplementing NMN. It's not a buzzing go get em type of energy. It's more like I just don't tire out like I was doing before. It's like there is always enough energy to do what I want and even then there is more if I need it. I work full time and I do regular overtime and I still work out and have time with my grandchildren. I just don't get really exhausted like I used to. When I lift weights, I lift a few reps of 100 kg's and within a minute or so I'm ready to lift again. Previously, lifting 100 kg's would leave me fatigued for up to 5 minutes. I assumed it was my ice-baths and perhaps the addition of Lugols Iodine, but the timing correlates more with the NMN, which I've been taking since January. Oh, and I've just remembered: (I'm a 56yr old cyclist) back in Feb and March I was feeling the cold nowhere near as much as usual on my bike, and the ice-baths and iodine didn't start until May and June respectively. I had plenty of energy anyway and workout with kettlebells nearly everyday alongside a healthy diet and time restricted feeding. I consider this protocol a kind of insurance. However, the heat tolerance aspect is one metric that is demonstrating to me that NMN is having an effect. I have a couple of comments to make and a question. I'm coming up on four months of taking NMN and, like I said in the comments of the previous video, I've come to a point I feel much better overall and it's now just an every day thing. It's the new normal. However, a few days ago something happened. I wouldn't say I feel physically better, it just my attitude took a leap upward. Like everything is about 25% better. Things that annoyed the crap out of me six months ago were definitely less so after starting NMN but now, as of a few days ago, even more so. Maybe it's just a cyclical thing like circadian rhythms. I guess we'll see. As far as people our age all being on medications. I agree. I live in a major resort town and it seems like everyone is healthy and strong. A big health problem here is a sports related injury. If I go back home to Kansas City, everyone there is on meds and has some type of health problem or two and they look old. My personal opinion is that cities are toxic. The lifestyle plus all the pollution is killing people. Finally, a question. I'm researching supplements such as fisetin and quercetin, even though I already take both. Do I really need both? It seems that fisetin is more effective by far and taking quercetin as well is redundant. Any thoughts? I tried backfilling methylation a different way - with choline. But strangely choline supplements brought on depression. You look very good for 58, healthy and vibrant. I just have one concern, your voice sounds hoarse and strained, almost like there is a pro-inflammatory issue. I sometimes experience this too when I have taken too many supplements and I believe this has to do with the sheer amount of substances that the liver has to break down which can cause production of toxic intermediates that generate pro-inflammatory free radicals. The liver must use methyl donor groups to break down phenolic compounds such as flavonoids and I believe there is a limit to how many methyl groups are available. Even flavonoids such as quercetin and fisetin can be pro-oxidant and pro-inflammatory. at high enough dosages. I read a study that found quercetin has cytotoxic effects on mouse cells when it was much higher than the equivalent of an average person taking only 10 mg a day, that is quite a low dose! This study also found that the mice had a shorter life span with higher quercetin levels. I can't find that study now but if I locate it again I will send you the ref citation.
