David Sinclair: Living to 120 and beyond | ABC Conversations Podcast

David Sinclair OAM is an Australian biologist based at Harvard University who is leading scientific fight against the inevitability of aging. Dr Sinclair believes aging is a disease, and we can find a cure for it. In the process, his lab has cured blindness in mice and has created tiny little brains out of human stem cells, which look like “little flesh-coloured peas”. Others like aubrey de grey think we have a 50% chance to extend our lives by 1 additional year each year within 15 years. Once at that point, we should pretty much live indefinitely unless we get hit by a car or something like that. 5 Reply Donna Oliver Donna Oliver 2 weeks ago A question in regard to fasting or time restricted feeding. We are told for example we need 4700mg of potassium per day, that is just one of many recommendations to keep our body running efficiently. With time restricted feeding how do we make sure we meet those goals? Is it possible that our body doesn't actually run on a 24 hour clock and we don't need this daily? Or does digestion itself dictate our 'daily' nutritional requirements. If you could live to the age of 300 then then what would be the problem with having kids and then REALLY focusing on your schooling in your mid 40s once the kids move out? It's interesting to talk about this subject, specifically aging and what the average human lifespan is, and what it could become if we only took better care of ourselves. The majority of the population, it seems, doesn't understand the impact of quality sleep, good diet, regular exercise (even 15-20 minutes a day), and sun protection on general health (and looks). I never took his medication but I changed my diet change my diet I went back to him 6 months later and he said wow did it ever work so good I've never seen the drug works so well and I told them I didn't take the medicine I changed my diet and he changed the subject. The initial testing came out negative so they told me there’s no way I had Lyme disease. When I finally got diagnosed after taking it into my own hands It had been 12 years. As a result I still have remaining paralysis in my face after being on antibiotics for five years in order to get it under control. It is funny how when we discover that the obvius is genius!!! My dad went almost to 100 here in Brasil following old school life style and tips he was a Karate and runner as well, and always use to tell us eat a little bit of everything with moderation and enough to feel it up both hands together not more than your stomach can handle. And not over or too much sex, sports, work and food just make in way that you enjoy enough to have a wish to do it again! The way to increase this is to get more individuals to live longer than the current average than die sooner. One way to do this is to persuade the members of the group to eat a healthy diet in moderation and to get regular exercise. I have been watching Dr. Sinclair's interview and bought his book “Why We Age and Why We Don’t Have To” very eye-opening and educative. In there, there's no point increasing lifespan without health span. The goal should be to live longer while staying young and healthy. Sinclair believes that in the foreseeable future, it's possible for humans to live beyond 120 or even 150, without the frailty, diseases and disability we now associate with old age. If you're seeing this dr sinclair can you tell me if you are basically consuming all your required calories intake in one go each day? If you are regularly fasting and only eat once, how do you eat possibly 1500 to 2000 or more calories on one meal or are you consuming a low calorie diet? Do you ever let yourself go when on holiday and eat local snacks ? I am 80 and often wonder what you will be saying in 10 or 20 years. Apparently your hip gives way and then you fall. Reason is that the western sugar/carb diet creates an acid environment resulting in a desperate search by the body for minerals to restore the acid/alkaline balance. Next week I will be starting an 11-day soft dry fast. I totally believe in dry fasting (not water fasting) now. I'll never do another water fast again. Drink my coffee in morning, followed by water, hot non caffeine tea and water in afternoon. Getting off my butt every hour walk around trying to get 4000 steps a day and more on days off. Whole non processed plants, fruit, egg and fish, beans. 65yo will see how it goes. but can tell fluid retention down, less joint pain already. This is for long haul. I just recently found your channel and really enjoy the content, especially when you dive into the details of Sinclair's comments and the studies. Since this one is about supplements, have you considered doing some videos on liposomal supplements? A lot of these longevity substances have very poor bioavailability. Using a liposomal delivery system dramatically increases the amount that gets into the bloodstream (almost all of it from what I've heard) and also facilitates entry into the cells where it is needed. I'm in the process of switching to liposomal (or phytosomes, which is very similar and does pretty much the same thing as liposomes) supplements where they are available for the stuff I take. One last thing, Metformin is mentioned in this video, and that is not typically available to people. You've also mentioned Rapamycin on your channel I believe as a longevity molecule. It turns out there are natural mimetics of those substances. On that reasoning I added ashwagandha, EGCG, and ginseng to my stack. Most of them are synergistic to the point of actually amplifying the effects of one another. The effects of Metformin and Aspirin, taken together, is more than the sum of each taken alone. The effects of Metformin and Resveratrol, taken together, is more than the sum of each taken alone. Metformin, Aspirin, Resveratrol and Quercetin are all synergistic, with many overlapping effects. I have taken all of these, and in similar doses (but 1 g Metformin per day) but only for 4 weeks, as a purge. I wouldn't want to risk doing this long-term. The only one I take every day is Metformin and that's because it's the only one of these compounds that has been proven to extend human life and has an excellent safety profile. On the other hand, one study on rats showed that Quercetin increased the occurrence of tumours. Starting Monday, I'll be taking the polyphenol, Pterostilbene together with NR as they seem to have a powerful anti-cancer effect when taken together. Pterostibene has 4X the bioavailability of Resveratrol so I'll only be taking 100 mg per day. I'll also be starting with sulforaphane and eating much more broccoli together with mustard seeds (trying looking that one up). When I was taking Resveratrol and Quercetin (my purge) I mixed them together with soya lecithin, on a slice of wholemeal bread. Both the lecithin and the wholemeal bread boost absorption, especially the lecithin, which has a strong affinty with water. I also mixed in 500 mg of VitC which I had in paste form, in capsules. The Vit C and lecithin help to protect the Quercetin, the lecithin in the stomach and the Vit C in the bloodstream, so it has more time in your system before being metabolised. I think this would work best as a complement to resveratrol. From what I've read they activate different areas of SIRT1. So using both gives more benefits. Also, I seein future videos you mention olive oil and avocados. I'm guessing for oleic acid? This activates SIRT6 (as well as SIRT1) which needs more research! SIRT1 gets the fame for now but SIRT6 may be just as important or possibly moreso for longevity! Can't wait to see that episode! I've always tried to include olive oil in my cooking. This was a very good video, and it confirms what I suspected, that resveratrol isn't a good senolytic. As I have said before, I take a total of 225mg daily, with 100mg being conventional resveratrol and 125mg being liposomal. This technically puts me in what you consider the medium range, but over half is liposomal, so presumably it reaches a lot more tissues and goes deeper into the cells. I also take quercetin every day, 250mg 3 times a day in phytosome form. Besides senescent cells there are benefits to quercetin like reduced inflammation. I know daily doses go as high as 1000mg, and I would guess that anything above that would put you in possible senolytic territory, which means I want to stay below that threshold. It also works as a zinc transporter which can boost your immune system if you're taking zinc (which I do). Recently I got some liposomal fisetin and began the Kirkland protocol, however I added liposomal 95% standardized OPC grape seed extract since pcc1 has shown evidence of being an excellent senolytic. I took these three days in a row, at 450mg fisetin and 600mg grape seed extract (I know the fisetin is below the Kirkland protocol, but my guess is liposomal delivery means you can go with less). However, after watching this video, I'm not sure how much benefit I can expect since I'm aged 45 and it looks like you need to be about 60 before you really need to kill off senescent cells. We definitely need more low dose fisetin studies. The liposomal pills I take are 150mg, so it would be nice to see what that dose would do. I am also not sure if doses are given as a bolus (all at once), or spread out over the day, and whether to use fat or black pepper to help with absorption. I take with a little fat (black olives or cocoa beans) I started donating plasma once a month (25% of plasma volume) at my local blood bank based Conboy studies, which among other things showed reduction of senescent cells in the brain. I took resveratrol every night with a high fat meal for a long peiod of time in doses between 600 - 1200mg and I've never been so sick in my life. My heart really worked hard and I got hypertension headaches & muscle weakness. I'm not a sensitive person at all and I've worked with supplements and taken alot for 10 years and never before got sick by supplements. I'm 31 and I'm a fully healthy individual who has never had high blood pressure before. I took resveratrol because nothing helped my tinnitus better than resveratrol for some reason. I'm not saying I believe it's necessarly bad just because I got side effects but definitly alot we dont know yet. Maybe Its better to take lower doses and less often as said in the video. This all happened 6 months ago and now when I took resveratrol 600mg just 5 days in a row again. I also follow Dr David Sinclair’s channel and read his books. By my own nature, I have never taken the megadoses which David Sinclair and other doctors recommend. I take the smaller doses, just to have some on board. I depend on a diet high in a wide variety of nutrient dense foods to do the heavy lifting for my health. Since you asked, I have one suggestion, that might improve your readership. Consider showing the full title, date, and with authors. Perhaps a direct link to the original study. For the reader it is important to sort out the wheat from the chafe. Perhaps the study is outdated by more recent studies. Perhaps the study is poorly designed. Such as a study with no placebo control. Is the study only on mice? On humans? In vitro? In vivo? What are the demographics of the study? Who, what age, country? How many persons enrolled and randomly controlled? Was the study performed over a period of a few months? Or, over a much longer term of many years? ETC ETC You do not need to point out any the details of each study. But when providing medical advice to the public, you do have a responsibility to cite the exact source of your study. Then the public can choose for themselves whether to look into a study more closely. Or the public might print out and bring your report to their medical provider. Without the studies cited, the doctor will not waste a moment to go any further. In the eyes of a professional medical doctor, your un-cited report is worth no more a mere rumor. Citing your sources clearly and fully takes up little space. This might raise your credibility immensely. 90% of “authentic” research scientists, and medical advisors to the public already publish in this way. I'm sure it took a lot of effort to pull it together and I hope your wife does fully recover. I have never thought of exercise as really promoting longevity, but it is likely a good tool. When it comes to autophagy, I'm deeply in the fasting camp, but I do exercise while fasting, especially if the fast is more than a day in length. Still really something to think about, thank you for your effort. I think exercise enables but doesn't guarantee anything. For example exercise isn't great for losing weight but it does build a base for good metabolic health. Additionally, about apoptosis, isn't that a frequent outcome of autophagy? In fact isn't that normally how muscles maintain? It isn't clear that apoptosis is a bad thing when it comes to muscles. I'm pretty sure that just a brisk walk can trigger some apoptosis in some muscles. Your goal is radically different than mine and I can't claim you are wrong any more than you can be sure your 14 day rest strategy will work long term. I kind of think you'll hit a point where you can't do as much but instead of finding a slight decline in muscles it will be much larger by then. The body often tends to use muscles differently than we expect. I also think it is difficult to come up with a set of HIIT exercises than hit all muscles. For what it is worth, I'm working on body weight exercises not just for muscle or leanness, but also to force my brain to learn new things. A bit of brain exercise, I hope. Still it really does seem that HIIT is very valuable and can save a lot of time, even though it can be painful or at least difficult. The examples that show exercise doesn't greatly promote longevity are rather extreme. For example body builders often only care about growing visible muscles and take extreme risks getting body fat very low. Other athletics were examples of people that train why beyond what most people would do. At best this is colorations and not sure conclusions can be really drawn from it. I used to do it every day, but I was losing too much weight, regardless of having increased my protein and fat intake. I also do intermittent fasting two low-carb meals a day. One meal will keep me too skinny, and it would probably affect my nutrition status (I was D and B12 deficient). This has worked great for me and has taken care of my prediabetic diagnosis. One thing I would say is that the detrimental effect of exogenous steroids on longevity coupled with the often terrible diets (for longevity) of pro bodybuilders would lead me to throw out the pro body builder paper from the analysis completely. All other factors you mention seem to point to body building as the most effective solution, even more than just HIIT. It makes sense if you think that often body-building routines involve high repetition sets (12+ reps) with short breaks (0.5->1.5 min) rests. This seems to satisfy that condition for rising and lowering oxygen levels. Also if you consider that the bodies response to high repetition resistance training is mass accrual, it follows logically that an attempt is made to keep as many living cells alive as possible. In fact, failure to complete a repetition often comes long before injury is possible due to the local production of heat shock proteins that inhibit the cells ability to work. My weekend walks usually last from 5 to 8 hours and that's exactly the type of exercise it gives me - without needing to run (which I don't enjoy). The ascents, which can be anything from 5 minutes to an hour, are intense - as intense as running - as my body is goes flat out to maintain a fast walking speed up a steep incline. Then there are periods of descending and level walking where I'm using very little energy. This is repeated many times throughout the day. The weight of my backpack helps too, expecially when my water bottles are still full. On weekdays, when I have less free time, I always go for a walk around the block once or twice. That involves a steep ascent too, where I walk as fast as I can, pushing my body. If, like me, you don't enjoy running or working out in a gym but live in a hilly area, there's a great alternative. Besides the physical exercise, I find it also helps me to unwind, think more clearly and it improves my mood. Yoga is a great stress reducer. Obesity is all too common today. Get off the couch. Get off the phone, ipad or video game. A variety of stretching and other exercises help with increased mobility.
